Dave Goodman (Devon Sawa), Sam Schecter (Jason Segel), and Jeff Davis (Michael Maronna) are best friends who have spent almost four years at Holden University scamming their way through college. During one exam scam in their final semester Dave discovers Angela (Jaime King, credited as James King), and asks her out while writing his phone number on her exam sheet. Ethan Dulles (who calls himself "Cool Ethan") (Jason Schwartzman), a classmate obsessed with Angela to the extent of collecting loose hairs and making a hair doll and having surveillance photos and a shrine to Angela setup in his dorm room, takes her exam question sheet after Angela leaves and uses it to repeatedly confront and then blackmail the guys into setting up a successful date with Angela for him in exchange for his silence on the matter. The guys set Ethan up in multiple situations in an attempt to convince Angela to like him, while Dave tells Sam during their work researching her that Angela is no more important to him as any other scam they've done. Ethan fails to attract her after frequent confrontations based on his delusional behavior, immaturity and ignorance of social norms.

In Mexico, a freelance vampire slayer, Derek Bliss (Jon Bon Jovi), is hired by an unknown client provided he build a team of slayers. Father Adam Guiteau (played by Tim Guinee in the first film) is shown to have been killed in this one. This team ends up including the vampiress Zoe, who is fighting her affliction with medication, Father Rodrigo, a helper who pretends to be a priest, and Sancho, a brave fourteen-year-old boy who aids Derek in killing some vampires at the beginning of the movie, in finding a large nest of "suckers" and their powerful leader, a vampire princess named Una (Arly Jover). She is seeking a legendary black crucifix: the Berziers Cross, the same crucifix used unsuccessfully in the first movie to perform a ritual which will enable vampires to walk in sunlight and be invulnerable.

Diana Guzman is a Brooklyn teenager whose hot temper gets her into trouble at school as she repeatedly starts fights with other students. The film is set in fictional Port Henry, Alaska, a town undergoing stress as the local economy switches from an industrial one based around the canning and paper industries towards a tourism and leisure based model. He now works as a handyman, particularly for Frannie and Lou, a lesbian couple who own the local resort hotel. Joe is friends with teenager Noelle De Angelo (Vanessa Martinez) who also works for Frankie and Lou. At an event which they are working, Noelle's mother Donna (Mary Elizabeth Mastrantonio), a lounge singer, breaks up with her live in boyfriend and asks Joe for help in moving. The two of them become close and eventually begin a romantic relationship. Meanwhile, Joe gets the chance to return to fishing when Frankie and Lou ask him to work a fishing boat which they have acquired as collateral from local fisherman Harmon. Donna has a strained relationship with her daughter Noelle, due mainly to Noelle's disapproval of her mother's peripatetic bohemian lifestyle. This is exacerbated when Donna begins dating Joe, who Noelle had had a crush on. At a bar Donna overhears the story of why Joe quit fishing: he had been involved in a deadly sinking which claimed the lives of all of his boatmates, including the brother of local bush pilot and small time criminal "Smilin Jack" Johannson (Kris Kristofferson).
