Policeman Philippe Jordan works in Marseille. He intercepts the delivery of a shipload of heroin by jumping out of a helicopter onto a speedboat and throws all drugs into the sea. Drug lord Mecacci is desperate to get rid of Jordan and arranges an incident which leads to Jordan's disciplinary transfer. Jordan continues to fight against the drug cartel after all. He finds a valuable witness named "Freddy, the chemist" but Mecacci has Freddy killed before he can testify. When Mecacci's henchmen also murder Jordan's old friend Francis Pierron, Jordan retaliates immediately. Now Mecacci tries to lure him into a deadly trap. After Jordan has outsmarted Mecacci's killers he confronts their boss. The time for the final showdown has come.

In mid-2000s, the illegal drug trade based in Kuala Lumpur is booming. The Malaysian authorities are called upon for assistance by their Indian counterparts. The Indian team sent to Kuala Lumpur is headed by Deputy Commissioner of Police D'Silva (Boman Irani), who targets the illegal drug operations of Singhania (Rajesh Khattar). Helping him along the way is Inspector Vishal Malik (Om Puri), an Indian officer from Interpol. Both of them know that in order to put an end to the illegal drug trade, they must capture "Don" (Shah Rukh Khan) who manages Singhania's business in Kuala Lumpur. Singhania was one of the two closest and most trusted men of Boris, a criminal who went underground and was presumed dead by the police. Boris's other lieutenant was a man called Vardhan, equally ruthless as Singhania, but his whereabouts are unknown. The police thinks that Singhania had killed Vardhan upon learning that Boris was about to hand over his drug business to Vardhan.

The film starts with ACP Yashvardhan IPS (John Abraham) in the hospital, recalling the accident that changed his life forever. Yash is a dutiful, responsible and stone-hearted police officer in the Narcotics Control Bureau whose only passion in life is crime-fighting. He has no family or loved ones and has been alone in life since he can remember. Everything changes when Yash meets Maya (Genelia D'Souza), an independent and lovely woman. Though the two get off to a rocky start, Maya warms up to him once she realises he is a police officer, and apologises for the misunderstandings they had. When Maya gets hit by a car in front of Yash, he rushes her to the hospital. With that, the two bond and start to meet and go out regularly. Picking up two or three years after where the original left off, narcotics officer Jimmy "Popeye" Doyle (Gene Hackman) is still searching for elusive drug kingpin Alain Charnier (Fernando Rey). Orders from his superiors send Doyle to Marseilles, France, to track down the criminal mastermind and bust his drug ring. Once in France, Doyle is met by Inspector Henri Barthélémy (Bernard Fresson), who resents his rude and crude crimefighting demeanor. Doyle then begins to find himself as a fish out of water in France, where he is matched with a language he cannot understand. Doyle is shown round the police station where he finds his desk is situated directly outside the toilets. He tells Barthélémy that he is not satisfied with this positioning and hopes it is not a joke at his expense. Barthélémy informs Doyle that he has read his personnel file and is aware of his reputation and especially hopes he has not brought a gun with him as it is strictly forbidden in France for visiting police officers from other countries to carry firearms.
