Farhan Qureshi (R.Madhavan) and Raju Rastogi (Sharman Joshi) are students at the Imperial College of Engineering (ICE), a highly regarded Indian engineering college. Virus is shown as an extremely strict person—so strict that he caused his own son's suicide by putting him under excess pressure. The star student of their class is Chatur Ramalingam (Omi Vaidya) nicknamed "Silencer", known for his incredible techniques of memorization but more notorious for his silent but malodorous flatulence in addition to his arrogant attitude. Catering for them and the other college students' needs is Man Mohan, nicknamed "Millimeter".

Ben Shaw (Joshua Jackson) and his new bride, Jane (Rachael Taylor), leave New York for Tokyo, Japan, where Ben has a job as a photographer. While traveling, Jane hits a girl in the middle of the wilderness, running over her. They find no body or even a trace of blood and decide to leave, thinking the victim was all right. They later start to find mysterious lights in their photos, which are later identified as spirit photography by Ben's assistant, Seiko Nakamura (Maya Hazen). Jane begins to have eerie dreams and visions as if they are trying to tell her something, and senses a haunting presence stalking them. Ben begins to complain of severe shoulder pain, and his friends begin to comment he's looking bent and hunched over, though the doctor can find no cause. Seiko takes Jane to her ex-boyfriend, Ritsuo (James Kyson Lee), whose career is to investigate paranormal activities, and he tells them that the lights are spirits, manifestations of intense emotions trying to communicate. At a subway station, Jane spots the ghostly presence of the girl she hit, causing her to believe that she killed the girl. Ben later also has a similar terrifying encounter. They go to a medium, Murase; however, Ben refuses to translate what Murase says, claiming he is a fraud.

In 2044, 25-year-old Joe works for a Kansas City crime syndicate as a "looper." Since technology in the future has made it almost impossible to successfully dispose of murdered bodies, the syndicates use time travel which was invented in 2074 and subsequently outlawed. Managed by a man sent from the future named Abe, loopers kill and dispose of victims sent back in time whose faces are hidden by a bag. They are paid with bars of silver strapped to the target. To prevent connections to the use of illegal time travel, when a looper is retired, his future self is sent back to 2044 as a target for his younger self, but with gold bars as a payment. This event is referred to as "closing the loop" and signals the end of a looper's contract. Failing to close the loop is punishable by execution.
