In 1607, the Susan Constant sails to the New World from England, carrying British settlers of the Virginia Company. On board are Captain John Smith and the voyage's leader Governor Ratcliffe, who seeks large amounts of gold in the New World to assure a strong position at the British court. Along the way, the Susan Constant is caught in a North Atlantic storm, and Smith saves a young, inexperienced crewmate named Thomas from drowning. In the Powhatan tribe in the New World, Pocahontas, daughter of Chief Powhatan, dreads being possibly wed to Kocoum, a brave warrior whom she sees as too "serious" for her free-spirited personality. Chief Powhatan gives Pocahontas her deceased mother's necklace as a present. Pocahontas, along with her friends, the gluttonous raccoon Meeko and hot-tempered hummingbird Flit, visit Grandmother Willow, a spiritual talking willow tree, and speaks of a possibly prophetic dream involving a spinning arrow, and her confusion regarding what her "path" in life should be. Grandmother Willow then alerts Pocahontas to the arriving British.

Ali G is the leader of Da West Staines Massiv, a fictional gang composed of a group of wannabe gangsters from Staines (a suburban town in north Surrey, to the west of London); their chief rivals are Da East Staines Massiv. Ali and Da West Staines Massiv are heartbroken to learn that their beloved local leisure centre (where they like to chill out and also where Ali teaches his "Keep it Real" classes) will be demolished by the local council. Ali and his friends decide to protest at this injustice. After he goes on a hunger strike and is spotted chained to some railings by the nefarious Chancellor of the Exchequer and Deputy Prime Minister David Carlton, he is drawn into a world of seedy political intrigue, as the Deputy Prime Minister tries to use Ali as a tool to destroy the Prime Minister's credibility. Ali is put forward as a candidate to be the next MP for Staines in a crucial by-election and manages to alienate most who cross his path, including feminists and the elderly. During a debate with his rival candidate, Ali tries to insult his rival by claiming that he "sucked off a horse". Unknown to Ali and the public, it turns out that the rival did indeed do such a thing, and trying to explain it away, he claims when he was out hunting with a friend he slipped, and his mouth landed on a horse's penis, which due to the mating season was erect. Ali then wins.

The film opens in 1968 at St. Anne's Academy, a California Roman Catholic school, where a young girl named Deloris Wilson is scolded by Sister Immaculata (Lois de Banzie) for wisecracking and disobedience. The setting then changes to the present day, where Deloris Van Cartier is a lounge singer in a 1960s-themed act called The Ronelles (a parody of The Ronettes), who sing at The Moonlite Lounge of the Nevada Club in Reno, Nevada, run by her boyfriend, the mobster Vince LaRocca. After Deloris walks in on Vince having his chauffeur Ernie executed for betrayal, Vince orders his two henchmen Joey and Willy to kill her as well. Deloris flees Vince's casino to the local police station where Lieutenant Eddie Souther suggests she testify against Vince if he can be arrested and tried, but for now, she should go into witness protection until the time comes.

Farhan Qureshi (R.Madhavan) and Raju Rastogi (Sharman Joshi) are students at the Imperial College of Engineering (ICE), a highly regarded Indian engineering college. Farhan's passion is wildlife photography, but he pursues an engineering degree to appease his father. He lives by the philosophy that one should pursue excellence, as when it is achieved, success follows. Virus is shown as an extremely strict person—so strict that he caused his own son's suicide by putting him under excess pressure. The star student of their class is Chatur Ramalingam (Omi Vaidya) nicknamed "Silencer", known for his incredible techniques of memorization but more notorious for his silent but malodorous flatulence in addition to his arrogant attitude. Catering for them and the other college students' needs is Man Mohan, nicknamed "Millimeter".
