The film follows Dr. Hess Green (Duane Jones), a wealthy black anthropologist who is doing research on the Myrthians, an ancient African nation of blood drinkers. One night, while staying in Green's lavish mansion --richly decorated with African art--, Green's unstable assistant George Meda (writer/director Gunn), attempts suicide. Green successfully talks him down, but later that night Meda attacks and stabs Green with a Myrthian ceremonial dagger, and then kills himself. Green, discovering the body, drinks Meda's blood and becomes a vampire endowed with immortality and a need for fresh blood. Though he steals several bags of blood from a doctor's office, he quickly finds that he needs fresh victims.

The film focuses on the exploits of the vengeful Dr. Gustav Niemann (Boris Karloff), who escapes from prison. He is helped by the hunchback Daniel (J. Carrol Naish), for whom he promises to create a new, beautiful body. The two murder Professor Lampini (George Zucco), a traveling showman, and take over his horror exhibit. To exact revenge on Bürgermeister Hussman (Sig Ruman), who had once caused his imprisonment, Niemann revives Count Dracula (John Carradine). Dracula seduces Hussmann's granddaughter-in-law Rita (Anne Gwynne) and kills Hussmann himself, but in a subsequent chase, Niemann disposes of Dracula's coffin, causing the vampire to perish in sunlight.

The Joker and the Penguin break out of Arkham Asylum and race each other to find a stash of stolen money hidden within a crypt in Gotham Cemetery after an inmate, who had hidden it, told them about it. The Joker is quickly intercepted by the Batman and is electrocuted when he falls into a river by his super-charged joy buzzers while the Penguin takes the opportunity to enter the cemetery himself. While searching through a crypt, the Penguin accidentally cuts his hand while using his umbrella-sword to open a coffin he hopes contains the money, finding a body instead. Blood from his hand drips on the corpse's heart, which brings it to life; it is none other than the vampire lord Count Dracula, his body having been moved from Transylvania to Gotham City after his "death." After attacking and turning a watchman at the cemetery into a vampire, Dracula hypnotizes the Penguin into becoming his non-vampiric servant so that he may lead him through Gotham to feed and guard his coffin during the day while he regains his strength.

A group of vampires look for the hidden resting place of Drake, also known as Dracula—the genetic "father" of their species. Thomas Hutter lives in the fictitious German city of Wisborg. His employer, Knock, sends Hutter to Transylvania to visit a new client named Count Orlok. Hutter entrusts his loving wife Ellen to his good friend Harding and Harding's sister Annie, before embarking on his long journey. Nearing his destination in the Carpathian mountains, Hutter stops at an inn for dinner. The locals become frightened by the mere mention of Orlok's name and discourage him from traveling to his castle at night, warning of a werewolf on the prowl. The next morning, Hutter takes a coach to a high mountain pass, but the coachmen decline to take him any further than the bridge as nightfall is approaching. A black-swathed coach appears after Hutter crosses the bridge and the coachman gestures for him to climb aboard. Hutter is welcomed at a castle by Count Orlok. When Hutter is eating dinner and accidentally cuts his thumb, Orlok tries to suck the blood out, but his repulsed guest pulls his hand away.
