Set in Sweden, the film is shown in flashbacks in the course of a trial in which a young woman with a half-hidden face, Anna Holm, is charged with murder. The events, as described by the witnesses, begin years before when aristocrat Torsten Barring hosts a party at a tavern. The guests include Vera, the faithless wife of noted plastic surgeon Gustaf Segert. When the tavern refuses to extend his credit, Torsten meets the proprietress Anna — whose face is badly scarred from a fire 22 years ago caused by her father. In the spring of 1918, a German U-boat stops a Dutch freighter on its way to Stockholm, Sweden during the final year of World War I. The Germans board the ship, identify one man as a Belgian spy, and take him prisoner. One of the passengers questioned is an attractive Swiss dress shop owner named Madeleine Goddard (Vivien Leigh) who is returning to Stockholm from a business trip to Paris, where she purchased dresses for her boutique. After returning to Stockholm, Madeleine, who is a German spy, meets with her contacts and provides information on Allied troop movements she acquired in Paris. The information is cleverly concealed in the gowns she purchased. While the Germans believe that she is one of their top spies, Madeleine is in fact a French double agent working to uncover the identity of the new German secret service leader in Stockholm.

The film focuses on a run-down boarding house in London, home to an assorted group of residents. Many of them cling precariously to their social positions with only one figure, the wealthy self-made businessman Mr Wright, being truly successful. The house is owned by the grasping Mrs Sharpe, who mistreats the maid, Stasia, a rehabilitated juvenile delinquent. The various members of the household are miserable and openly sneering and rude towards each other, the one exception being the respect shown by all to the powerful Mr Wright. In the case of one couple, Major Tomkin and his wife, this involves pressuring their daughter Vivian to marry Wright in spite of her obvious horror at the idea.

In 1593 London, William Shakespeare is a sometime player in the Lord Chamberlain's Men and poor playwright for Philip Henslowe, owner of The Rose Theatre. Shakespeare is working on a new comedy, Romeo and Ethel, the Pirate's Daughter. Suffering from writer's block, he has barely begun the play, but starts auditioning players. Viola de Lesseps, the daughter of a wealthy merchant, who has seen Shakespeare's plays at court, disguises herself as "Thomas Kent" to audition, then runs away. Shakespeare pursues Kent to Viola's house and leaves a note with the nurse, asking Thomas Kent to begin rehearsals at the Rose. He sneaks into the house with the minstrels playing that night at the ball, where her parents are arranging her betrothal to Lord Wessex, an impoverished aristocrat. While dancing with Viola, Shakespeare is struck speechless, and after being forcibly ejected by Wessex, uses Thomas Kent as a go-between to woo her. Wessex also asks Will's name, to which he replies that he is Christopher Marlowe.

In 1945, as a truckload of war survivors stops in front of an Amsterdam factory at the end of World War II, Otto Frank (Joseph Schildkraut) gets out and walks inside. After climbing the stairs to a deserted garret, Otto finds a girl's discarded glove and sobs, then is joined and comforted by Miep Gies (Dodie Heath) and Mr. Kraler (Douglas Spencer), factory workers who shielded him from the Nazis. After stating that he is now all alone, Otto begins to search for the diary written by his youngest daughter, Anne. Miep promptly retrieves the journal for Otto, and he receives solace reading the words written by his young daughter three years earlier.
