Roshan (Abhishek Bachchan) accompanies his dying grandmother Annapurna (Waheeda Rahman) to their ancestral property in crowded Delhi. Roshan is initially stunned by the mad rush of neighbours: Ali Baig (Rishi Kapoor) the renaissance man, feuding brothers Madangopal (Om Puri) and Jaigopal (Pavan Malhotra), their wives and families, Mamdu (Deepak Dobriyal) the halwai, Gobar (Atul Kulkarni) the simpleton, Sethji and many others. Roshan eventually warms to the place and wholeheartedly embraces the sense of community. He accompanies his grandmother to the Ramleelas, hangs out at Mamdu's sweet stall, plays with the children, and gradually becomes steeped in the culture.

A young, struggling British filmmaker Sue McKinley (Alice Patten) comes across the diary of her grandfather, Mr. McKinley (Steven Mackintosh), who served as a jailer in the Imperial Police during the Indian independence movement. Through the diary, she learns about the story of five freedom fighters who were active in the movement: Chandrasekhar Azad, Bhagat Singh, Shivaram Rajguru, Ashfaqulla Khan, and Ram Prasad Bismil. McKinley, in his diary, states that he had met two type of people in his life: the first one... who died without uttering a sound and the second kind ... who died with lots of anguish.. crying over their deaths... McKinley reveals that it was then that he met with the third kind.

The film starts in the 1960 Summer Olympics in Rome, where a coach says "Bhaag Milkha Bhaag!" The story is taken back to the memories of the childhood days of a young boy which haunted him, resulting in him dropping to fourth. Partition of India in 1947 caused chaos which resulted in mass religious violence in Punjab in British India, killing the parents of Milkha Singh (Farhan Akhtar). He reaches Delhi and later meets his sister there. Living in impoverished refugee camps, Milkha soon makes friends and survives by stealing along with his friends. He falls in love with Biro (Sonam Kapoor), but she asks him to live a life of honesty.

In 2008, Deepak Kumar (Saif Ali Khan) is at an interview for the teacher's post at an affluent school. The interviewers turn him down when they discover his low-caste roots. A young woman (Urmila Matondkar), home alone, is watching the news of a serial killer on television when the doorbell rings. The woman sees through the peephole that it is a young man, who later identifies himself as Sameer A. Purnavale (Manoj Bajpai), claiming to be a business partner of Mr. Malhotra. She is reluctant to open the door, due to the news broadcast of a killer on the loose. The woman claims that the house doesn't belong to Mr. Malhotra but to Mr. Gupta. However the man believing that there has been a mix up, persistently keeps ringing the doorbell even after the woman tells him that she won't let him in. To scare him away the woman lies and tells the man that her husband is sleeping upstairs. The man then claims that he hears a noise upstairs so that if her husband is awake can he talk to him. Upon hearing this and a subsequent noise from within the house, the woman is spooked and runs outside.

A gang of heavily armed men scour and finally narrow down on a house in Wasseypur. They surround the house and unleash a wave of bullets and grenades on it with the intention of killing the family inside it. After heavy firing on the house, they retreat from the crime scene in a vehicle, convinced they have killed everyone within. 'Don' begins, true to its title, as the story of one of the most powerful men in the business of crime. In spite of being on Interpol's Most Wanted list, Don remains elusive to the police. Besides the police, Don makes a few other enemies through his merciless approach to running his organization. Notably, Don kills one of his own men, Ramesh, when Ramesh decides to leave the business. This introduces Don to two new enemies, Kamini (Helen), Ramesh's fiancee, and Roma, (Zeenat Aman) Ramesh's sister. When Kamini seduces Don and attempts to have the police arrest him, her plan backfires as Don outsmarts her and the police in his escape. In the process, Kamini is killed. A shattered, revenge-seeking Roma gets her hair cut short, trains in judo and karate, then enters Don's gang after deceiving them into thinking that she too is on the wrong side of the law. Don is impressed with her fighting skills and allows her to work for him without suspecting any ulterior motive.
