In 1931, China is embroiled in a civil war. Friends of British Captain Donald "Doc" Harvey (Clive Brook) envy him because the fabulously notorious Shanghai Lily (Marlene Dietrich) is a fellow passenger on the express train from Beiping to Shanghai. Since the name means nothing to him, they inform him that she is a "coaster" or "woman who lives by her wits along the China coast" – in other words, a courtesan. On the journey, Harvey encounters Lily, who turns out to be his former lover, Magdalen. Five years earlier, she had played a trick on Harvey to gauge his love for her, but it backfired and he left her. She frankly informs him that, in the interim, "It took more than one man to change my name to Shanghai Lily." When Lily makes it clear that she still cares deeply for him, it becomes apparent that his feelings also have not changed, and he shows her the watch she gave him with her photograph still in it.

Sophia Frederica (Dietrich) is the daughter of a minor German prince and an ambitious mother. She is brought to Russia by Count Alexei (Lodge) at the behest of Empress Elizabeth (Dresser) to marry her nephew, Grand Duke Peter (played as a half-wit by Jaffe in his film debut). In 1928 Hollywood, director Leo Andreyev (William Powell) looks through photographs for actors for his next movie. When he comes to the picture of an aged Sergius Alexander (Emil Jannings), he pauses, then tells his assistant (Jack Raymond) to cast the man. Sergius shows up at the Eureka Studio with a horde of other extras and is issued a general's uniform. As he is dressing, another actor complains that his continual head twitching is distracting. Sergius apologizes and explains that it is the result of a great shock he once experienced.

A Russian defector lands a jet fighter aircraft on an American airstrip. The base commander, Air Force Colonel Jim Shannon (John Wayne) is surprised to find that the pilot is an attractive woman, Lieutenant Anna Marladovna (Janet Leigh). When she asks for asylum, but refuses to disclose any military information, Shannon is assigned to seduce her. They fall in love. The Town presents an idealized vision of American life, shown in microcosm by Madison, Indiana. The diversity of the town's ethnic origins is highlighted, noting the Czech, Dutch, German, and Italian communities, some of whom were immigrants or children of immigrants. Schools are shown to be free and open to all, as are libraries and swimming pools. The press is depicted as free. Some people are shown who don't like the current administration in Washington, and don't like the newspaper's policy, but the newspaper prints their complaint. Trials are conducted in front of a jury, for all the world to see. The prosecutor serves as the judge's opponent in the last election, and even said he was unfit for office, but they work together anyway. Everyone had the right to vote, from the store keeper, to the attorney himself. In a democracy, the only thing that is secret is the ballot.

Englishman A. J. Fothergill (Robert Donat) is recruited by Colonel Forrester (Laurence Hanray) to spy on Russia for the British government because he can speak the language fluently. As "Peter Ouranoff", he infiltrates a revolutionary group led by Axelstein (Basil Gill). The radicals try to blow up General Gregor Vladinoff (Herbert Lomas), the father of Alexandra (Marlene Dietrich). When the attempt fails, the would-be assassin is shot, but manages to reach Peter's apartment, where he dies. For his inadvertent involvement, Peter is sent to Siberia.
