The story deals with the joys and tribulations of a travelling minstrel troupe known as the Merry Meadow Minstrels. Al Jolson plays as a blackface endman while Lowell Sherman plays as the interlocutor. Hobart Bosworth plays as the owner of the show, while his daughter, played by Lois Moran, serves as Al Jolson's love interest in the picture. Sherman's character, however, is also in love with Moran's. The show is in a miserable state until Jolson entertains a sheriff and manages to convince him to invest in the show. The show becomes very successful thanks to this investment and Jolson is eventually able to visit his mother. Some time after he returns, he tells Moran that he loves her and this causes Sherman to become jealous. After a heated argument between Jolson and Sherman over Moran, a character played by Mitchell Lewis, who is upset because he was caught cheating at cards, puts real bullets in Jolson's stage gun. Since Jolson pretends to shoot Sherman in the minstrel show act, Lewis knows that this will result in Sherman's death and that Jolson will be blamed for the murder. After Sherman is shot, Jolson is arrested but manages to escape and take a freight train out of town. Eventually, Lewis confesses to the crime and Jolson is thereby proven to be innocent.

Youngster Frankie (Gene Reynolds) and his small gang commit petty crimes in their New York City tenement neighborhood, such as stealing bicycles and taking money from other boys. In the early days of World War II, Cohan comes out of retirement to star as President Roosevelt in the Rodgers and Hart musical I'd Rather Be Right. On the first night, he is summoned to meet the President at the White House, who presents him with a Congressional Gold Medal (in fact, this happened several years previously). Cohan is overcome and chats with Roosevelt, recalling his early days on the stage. The film flashes back to his supposed birth on July 4, whilst his father is performing on the vaudeville stage.

Connie Ward (Ann Rutherford) is a young woman who on the spur of the moment marries Bill Abbott (George Montgomery), a trumpet player in Gene Morrison's (Glenn Miller) swing band (Miller's character was given a name with initials that matched Miller's so that the band could use their monogrammed stainless-steel music stands). She soon finds herself at odds with the cattiness and petty jealousies of the other band members' spouses, as they accompany their husbands on their cross-country train tour. Her discomfort is exacerbated by a flirtation between Abbott and Jaynie (Lynn Bari), the band's female vocalist. When Ward eventually walks out on Abbott, their split releases so many other tensions among the musicians and their wives, that leader Morrison is forced to break up the orchestra. Ward and the band's pianist Sinjin (Cesar Romero) then work behind the scenes to reunite the band, which also produces a reconciliation between Ward and Abbott (with additional help from Connie's father (Grant Mitchell)).
