Lamont Cranston is an amateur criminologist and detective, who hosts a daily radio program sponsored by the Daily Classic newspaper. He has developed a friendly but occasionally terse feud with Police Commissioner Weston. Cranston complains to his managing editor, Edward Heath, about his incompetent new assistant, Phoebe Lane. Heath advises him that because she is the publisher's niece, she cannot be fired. During his radio broadcast, Phoebe gives Cranston a note that the Metropolitan Theatre is to be robbed at eight o'clock. Afterwards, he learns she got the information from a man she met in a café. Hiro Hamada is a 14-year-old robotics genius in the futuristic fictional city of San Fransokyo. Raised by his aunt Cass and older brother Tadashi after the death of his parents, he spends his time participating in illegal robot fights. To redirect Hiro, Tadashi takes him to the robotics center at his university, where Hiro meets Tadashi's friends: GoGo, Wasabi, Honey Lemon, and Fred, as well as Baymax, the inflatable healthcare companion robot Tadashi created. To enroll in the school, Hiro signs up for the school's science fair and presents microbots: swarms of tiny robots that can link together in any arrangement imaginable. Professor Callaghan, the head of the university's robotics program, is impressed, and grants Hiro entrance to the university. Alastair Krei, renowned entrepreneur and president of Krei Tech, offers to buy the microbots, but Hiro follows Callaghan's warnings about Krei's questionable business practices and declines to sell them. When Tadashi and Hiro later arrive at the university to find it engulfed in flames, Tadashi rushes in to rescue Callaghan, but the building explodes moments later, killing him.

The film opens with a slight introduction from the main hero of the film, Underdog who introduces the audience to himself, his arch-nemesis Simon Barsinister, and Barsinister's henchman Cad. The film then enters into Capital City Town Hall, where a beagle on the police bomb squad sets off a false alarm after smelling a large ham (in some boxes near where the mayor was giving a speech). After being shunned by his fellow police canines, the beagle leaves in shame and is abducted off the street by Cad, who takes him to Simon's lab. Simon is bitter against the mayor, who has turned down his proposal for a type of genetic experimenting for the city, and plans to prove him wrong by testing his new serum on the beagle. However, Underdog escapes and runs amok in the lab, causing a fire and accidentally becoming exposed to a variety of DNA concoctions and chemicals that Simon developed, which modifies his DNA and gives him the strength to break through the metal door to the lab. After witnessing his work's "success" Simon is stunned when a burning monitor falls onto him, and Cad rushes to his aid.

In the metropolis of Champion City, the would-be superhero team of Mr. Furious (Ben Stiller), The Shoveler (William H. Macy), and The Blue Raja (Hank Azaria) attempt to make a name for themselves, but their suspect skills make them ineffective, and they find themselves upstaged by the city's most successful superhero, Captain Amazing (Greg Kinnear). However, Amazing is finding that his effectiveness at fighting crime has practically made his job obsolete, and without any worthy adversaries remaining (some of them are either dead, in exile, or still in jail), his corporate sponsors are beginning to pull their funding. To create a need for himself, Amazing uses his alter ego, billionaire lawyer Lance Hunt, to argue for the release of insane supervillain Casanova Frankenstein (Geoffrey Rush). The plan backfires; once reunited with Tony P (Eddie Izzard) and his Disco Boys, Casanova Frankenstein blows up the insane asylum, captures Amazing, and prepares to unleash a doomsday device: the "Psycho-frakulator", which lethally bends reality. Mr. Furious, while spying on Casanova Frankenstein's mansion, discovers Amazing's capture and informs the others.

Five-year-old Krishna Mehra undergoes an intelligence quotient test by a professor, who suspects that he has superpowers.
