Daanish (Dhanush), born mute in rural Igatpuri, loves films. He acts out scenes before his class, amazing his peers and teacher, and spends all his time in the local Paradise theatre. He even tries to board a Mumbai bus to go become a film star but is pulled off by his mother's friends. Scientist Rohit Mehra lives with his son Krishna "Krrish" Mehra (both played by Hrithik Roshan) in Mumbai. Krishna is married to Priya (Priyanka Chopra) who works as a journalist for Aaj Tak. Rohit works in a research institute for the Indian government and Krishna, while moon lighting as the superhero Krrish, is regularly fired from various day jobs due to lack of attendance. Rohit is researching on a device that will bring life to dead tissues by the use of solar energy. Despite several attempts, he fails to complete the experiment due to the high intensity of sun rays. On a usual day, Krishna loses his job from the security wing of a company when he hears news about an aeroplane whose front tyres are stuck due to some malfunctioning which creates high risk, even if landed, it may explode and so will the passengers inside. Krishna immediately changes himself to Krrish and saves the plane from accident by holding the component to make tyres free to roam.

During the 1947 partition, the Lahore based family of Rajveer (Rishi Kapoor) and Amrit (Rekha) has to flee Pakistan and settle in Amritsar, Punjab. In the house that they get to stay, Amrit finds an abandoned baby boy of the Muslim family who owned the house but fled to Pakistan because of communal riots. Amrit raises the boy as her own and he grows up to be Ishaan (Luv Sinha). During a summer camp visit to Kashmir, Ishaan falls in love with Chandni (Ferena Wazeir). When he goes to her house to ask her hand in marriage, her father (Deep Dhillon) and uncle (Ahmed Khan) tell him to forget her as they are against her marrying a Sikh boy. When Amrit and Rajveer come to know about this they finally declare the truth to Ishaan that he is a Muslim in reality and not their own child. He doesn't believe them and Chandni's parents refuse to believe it without proof. The old couple then decide to track Ishaan's real parents down and also succeed. Ishaan's real mother, Benazir (Hema Malini) comes down with his real father (Javed Sheikh) to take back custody of a now grown up Ishaan. Chandni's parents immediately agree to the marriage when Ishaan's real parents visit their house. What complications arise when Ishaan's parents start making plans to take back Ishaan and his bride back to Pakistan and how they are handled by the principal characters forms the rest of the plot.

The movie starts with a flashback, in front of a church, in 1980 when the four main characters were children. This scene the beginning of their friendship. Thomas Philip (a.k.a. Thomas Kutty) loses money to a scam artist, who responds to his complaints with violence. He is rescued by the other three who respond to his calls of distress. They then invite Thomas Kutty to join their group who, above all, vow never to cheat on one another. The scene is transitioned by the quote "Thomas kutty Vittodaa!" (Thomas kutty, time to run!) once the scam artist brings his friends to return the beating he got.

Om Prakash Makhija (Shah Rukh Khan) is a junior artist in the 1970s Hindi film industry, in love with film star Shanti Priya (Deepika Padukone). One evening, Om views the premiere of Shanti's film Dream Girl, posing as Manoj Kumar. Later that night, Om becomes drunk and describes his vision of himself as a famous, wealthy film star winning the Filmfare Award for Best Actor to his friend Pappu (Shreyas Talpade) and some local children. At the shooting of a later film, Om rescues Shanti from a fire scene where of the fire has escaped control, and they become friends. One day, after following Shanti, he overhears her and producer Mukesh Mehra's (Arjun Rampal) conversation, that Mukesh and Shanti have married in secret and Shanti is pregnant and expects a traditional wedding. Mukesh pretends to be happy with the news, and invites Shanti at the set of their soon-to-be-made film Om Shanti Om. Mukesh promises to cancel the film, reveal their marriage to the public, and have a grand wedding on the set; but instead traps Shanti on the set and sets it on fire to prevent a financial loss and to protect his career. Shanti tearfully begs Mukesh to release her but he leaves anyway.
