Synopsis
Young actor Vikram Jaisingh (Farhan Akhtar) arrives in Mumbai to make it as a Bollywood film star with the help of Abhimanyu (Arjun Mathur), an actor friend from his hometown, and their mutual friend Sameer, who works in a studio props department.Actors

At the beginning of the film, Kabir (Abhay Deol) proposes to Natasha (Kalki Koechlin). Their families meet at the engagement party where Natasha learns that Kabir is planning a three-week bachelor road trip to Spain with his school friends Imraan (Farhan Akhtar) and Arjun (Hrithik Roshan). Kabir explains that the three have a long-standing pact, and that during the road trip, each of them will have to pick an adventure sport, which all three will try together. Initially, Arjun is reluctant to take trip as he does not want lose his pay for missing work. It is later revealed that Arjun's ex-girlfriend left him due to his workaholism & obsesion with money. Imraan has an additional personal agenda on the trip: to find his biological father Salman Habib (Naseeruddin Shah), an artist in Spain.

Kamal Mehra (Anil Kapoor) is a businessman who owns Ayka, a company on the verge of bankruptcy. Karthik (Farhan Akhtar) is an introvert who lacks confidence and feels trapped in his average job at a construction company. Om Prakash Makhija (Shah Rukh Khan) is a junior artist in the 1970s Hindi film industry, in love with film star Shanti Priya (Deepika Padukone). One evening, Om views the premiere of Shanti's film Dream Girl, posing as Manoj Kumar. Later that night, Om becomes drunk and describes his vision of himself as a famous, wealthy film star winning the Filmfare Award for Best Actor to his friend Pappu (Shreyas Talpade) and some local children. At the shooting of a later film, Om rescues Shanti from a fire scene where of the fire has escaped control, and they become friends. One day, after following Shanti, he overhears her and producer Mukesh Mehra's (Arjun Rampal) conversation, that Mukesh and Shanti have married in secret and Shanti is pregnant and expects a traditional wedding. Mukesh pretends to be happy with the news, and invites Shanti at the set of their soon-to-be-made film Om Shanti Om. Mukesh promises to cancel the film, reveal their marriage to the public, and have a grand wedding on the set; but instead traps Shanti on the set and sets it on fire to prevent a financial loss and to protect his career. Shanti tearfully begs Mukesh to release her but he leaves anyway.

Famed playwright Sidney Bruhl (Caine) debuts the latest in a series of Broadway flops and returns to his opulent Long Island home and his wife, Myra (Cannon).
