The film stars Vardalos, John Corbett, Andrea Martin, Ian Gomez and Elena Kampouris. It is the sequel to My Big Fat Greek Wedding. Filming began in late-May 2015 in Toronto. The film is scheduled for a March 25, 2016 release.

Fotoula "Toula" Portokalos (Nia Vardalos) is going through an early midlife crisis. At thirty, having had her childhood, adolescence and adult life plagued by bad luck, she feels that she is the only woman in her family who has "failed" at being a typical Greek girl. Her family expects her to be more like her 33-year-old sister Athena (Stavroula Logothettis) and marry a Greek boy, make Greek babies, and "feed everyone until the day she dies." Instead, Toula is stuck working in the family restaurant in Chicago, "Dancing Zorba's." Frumpy and cynical, she fears that she is doomed to be stuck with her life as it is and always has been. It tells the World War II era story of a cynical, successful businessman named Domenico (Mastroianni), who, after meeting a naive country girl, Filumena (Loren), one night in a Neapolitan brothel, keeps frequenting her for years in an on-off relationship (as she continues working as a prostitute). He eventually takes her in his house as a semi-official mistress under the pretense that she take care of his ailing, senile mother. After having fallen for a younger, prettier girl and having planned to marry her, he finds himself cornered when Filumena feigns illness and "on her deathbed", asks to be married to him. Thinking she'll be dead in a matter of hours and that the 'marriage' won't even be registered, he agrees. After having been proclaimed his legal bride, the shrewd and resourceful Filumena drops the charade and reveals to have put up the show for the one child she bore from him (she gave birth to three sons but Domenico always maintained to have fathered none). Domenico tries to cajole her into telling him which one is his but she stalwartly refuses, telling him that sons can't be picked and chosen and that he has to be the father of all three. When word reaches Jackie O'Shea (Ian Bannen) and Michael O'Sullivan (David Kelly), two elderly best friends, that someone in their tiny Irish village of 52 people in Tulaigh Mhór (Tullymore) has won the Irish National Lottery's Lotto game, they, along with Jackie's wife Annie (Fionnula Flanagan), plot to discover the identity of the winner. They obtain a list of lottery customers from Mrs. Kennedy (Maura O'Malley) at the post office, and invite the potential winners to a chicken dinner, where they attempt to get the winner to reveal his or herself. After everyone has left and they are no closer to an answer, Annie realizes that one person did not come to the dinner, so Jackie pays a late-night visit to the only absentee: the reclusive Ned Devine (Jimmy Keogh). He finds Ned in his home in front of the TV, still holding the ticket in his hand, a smile on his face and dead from shock. That same night, Jackie has a dream that the deceased Ned wants to share the winnings with his friends, as he has no family to claim the ticket. Jackie wakes up after the dream, and before dawn, he and Michael return to Ned's house to gather Ned's personal information so they can claim the winnings for themselves.

Boxer and amateur pilot Joe Pendleton (Robert Montgomery), affectionately known as "the Flying Pug", flies his small aircraft to his next fight in New York City, but crashes when a control cable severs. His soul is "rescued" by 7013 (Edward Everett Horton), an officious angel who assumed that Joe could not have survived. Joe's manager, Max "Pop" Corkle (James Gleason), has his body cremated. In the afterlife, the records show his death was a mistake; he was supposed to live for 50 more years. The angel's superior, Mr. Jordan (Claude Rains), confirms this, but since there is no more body, Joe will have to take over a newly dead corpse. Mr. Jordan explains that a body is just something that is worn, like an overcoat; inside, Joe will still be himself. Joe insists that it be someone in good physical shape, because he wants to continue his boxing career. Joe keeps saying the body they find "has to be in the pink".
