The film tells the forgotten story of Andrée Heuschling, also known as Catherine Hessling, who was the last model of impressionist painter Pierre-Auguste Renoir and the first actress in the films of his son, the film director Jean Renoir. Andree was the link between two famous and widely acclaimed artists, a father and son. While the father is at the end his brilliant career, the son is still searching for himself, his great career as one of the most celebrated movie directors having not yet begun. Vincent van Gogh's obsessive devotion to his art engulfs, consumes, and finally destroys him. The apostate religious leaders do not like his zeal for God and they frown on his social activism and care for the poor in a coal mining town. He returns home to his father's house where he is rejected by a woman he obsessively loves, takes up with a prostitute who leaves because he is too poor, and discovers painting, which he pursues while agonizing that his vision exceeds his ability to execute. His brother, Theo van Gogh, provides financial and moral support, while Vincent lives off and on with the critical Paul Gauguin. Vincent begins experiencing hallucinations and seizures and voluntarily commits himself to a mental institution. He signs himself out, and with Theo's help, returns to a rural area to paint, where he ultimately shoots himself in despair of never being able to put what he sees on canvas.

Griet (Scarlett Johansson) is a shy girl living in the Dutch Republic in 1665. Her father, a Delftware painter, has recently gone blind, rendering him unable to work and putting his family in a precarious financial situation. To help matters, Griet is sent to work as a maid in the household of famed painter Johannes Vermeer (Colin Firth). Griet works hard, almost wordlessly, in the lowest position in a harsh hierarchy, doing her best despite spiteful treatment by one of Vermeer's children. Le film, comme le titre l'indique, est une biographie du peintre hollandais du XVII siècle, qualifié de « plus grand peintre de tous les temps » dans le texte qui défile à l'écran en guise de préambule. Rembrandt, au moment où l'histoire commence, est un peintre adulé, sollicité par les plus riches, mais dont le plus grand plaisir consiste à réaliser des portraits de son épouse Saskia. Celle-ci, cependant, tombe brusquement malade et meurt, laissant le peintre désemparé. Plutôt que d'assister aux funérailles de sa femme, il tâche de peindre d'elle au moins un dernier portrait, tant que son image ne s'est pas encore tout à fait éteinte dans son souvenir ; à partir de cet événement tragique, la vie du peintre, progressivement, va se désagréger…
