After drifting apart emotionally over the years, two single siblings — Wendy (Linney) and Jon (Hoffman) — band together to care for their estranged, elderly father, Lenny (Philip Bosco), who is rapidly slipping into dementia. Wendy and Jon first travel to Sun City, Arizona to attend the funeral of their father's girlfriend of 20 years. American Splendor retrace la véritable histoire du scénariste de comics Harvey Pekar. Au début du film, celui-ci mène une existence plutôt morose entre son travail de documentaliste, quelques conversations sur tout et rien avec ses collègues et quelques connaissances, et sa passion pour le jazz, qu'il assouvit en recherchant des pièces rares dans les garage sales (équivalent américain des vide-greniers) ou les magasins d'occasion. C'est ainsi qu'il fait la connaissance du célèbre dessinateur de bande dessinée Robert Crumb, alors inconnu, qui est lui aussi collectionneur. Plusieurs années plus tard, alors que Crumb est devenu célèbre, Harvey lui soumet les story-boards d'un projet de bande dessinée basée sur sa vie, lui proposant d'en assurer le dessin. Crumb accepte, et contre toute attente, cette série intitulée American Splendor relatant une vie simple, banale, obtient un grand succès. Harvey Pekar acquiert ainsi une certaine célébrité aux États-Unis, dont les conséquences sur sa vie quotidienne sont présentées dans le film. En particulier, American Splendor lui permet de rencontrer sa future femme Joyce Brabner, qui écrit elle aussi des comics, et avec qui il va réaliser d'autres histoires autobiographiques, également intégrées en partie dans le film (notamment Our Cancer Year).

Constructed as a series of chapters that take place at a turning point in each character's life, the story moves from seven years in the past to three years to two and finally arrives in the present day. Cal Weaver (Steve Carell) is a middle-aged man who learns that his wife Emily (Julianne Moore) cheated on him with a co-worker, David Lindhagen (Kevin Bacon), and that she wants a divorce. After moving into his own apartment, Cal goes to a bar night after night, talking loudly about his divorce, until he attracts the attention of a young man named Jacob Palmer (Ryan Gosling), a womanizer who beds women each night, although a young woman named Hannah (Emma Stone) recently had rejected his advances. Jacob takes pity on Cal, and offers to teach him how to pick up women. Using Jacob's teachings, Cal seduces Kate (Marisa Tomei) at the bar. After this encounter, Cal manages to successfully seduce other women in the bar. He sees Emily again at their son Robbie's (Jonah Bobo) parent-teacher conference. The interaction goes well until they discover that Robbie's teacher is Kate, who reveals to Emily that she and Cal slept together. Cal then confesses to sleeping with many women. Emily leaves in disgust and begins actively dating David. Meanwhile, Hannah, a recent law school graduate, is expecting her boyfriend, Richard (Josh Groban) to propose marriage while they celebrate her passing the bar exam, but he does not, instead, offering her a position at his law firm. Offended and hurt, Hannah returns to the bar where she originally rejected Jacob's advances and kisses Jacob passionately. The two return to Jacob's home to hook up, but end up talking to each other all night and making a connection. Jacob starts a relationship with Hannah, and he becomes distant from Cal.

Laurel and Audrey are identical twins. Although they look the same on the outside, their personalities are very different. When they get in a severe car crash together, Laurel is taken to the hospital and is informed that her sister has died. Suffering from post-traumatic amnesia, Laurel doesn't remember being who she is at first. The morning of her sister's funeral, she remembers who she is but decides to keep everyone thinking that it was she who had died and not Audrey. She then flies back to the city where Audrey used to live. There, she meets her sister's tenant Basel (Jake Johnson) who is confused about her sudden change of personality, because Audrey was never nice to him before. Living her life posing as her sister, she learns that Audrey had a boyfriend who is married, but broke up with him before the accident. Laurel continues to spend more time with Basel and Audrey's best friend Claudia, who is suspicious about Audrey's change of behavior. Laurel begins a relationship with Basel, and they fall in love. When she finally feels that she is adapting to her new life, she accidentally tells her boss that Charles is her ex-boyfriend, not knowing that Charles is actually her boss's husband, and gets fired immediately. Shortly after this, Basel proposes to Laurel saying that he loved her since he first met her. Laurel, unable to continue deceiving everyone, decides to tell Basel the truth, but he is devastated and breaks up with her.
