Om Prakash Makhija (Shah Rukh Khan) is a junior artist in the 1970s Hindi film industry, in love with film star Shanti Priya (Deepika Padukone). One evening, Om views the premiere of Shanti's film Dream Girl, posing as Manoj Kumar. Later that night, Om becomes drunk and describes his vision of himself as a famous, wealthy film star winning the Filmfare Award for Best Actor to his friend Pappu (Shreyas Talpade) and some local children. At the shooting of a later film, Om rescues Shanti from a fire scene where of the fire has escaped control, and they become friends. One day, after following Shanti, he overhears her and producer Mukesh Mehra's (Arjun Rampal) conversation, that Mukesh and Shanti have married in secret and Shanti is pregnant and expects a traditional wedding. Mukesh pretends to be happy with the news, and invites Shanti at the set of their soon-to-be-made film Om Shanti Om. Mukesh promises to cancel the film, reveal their marriage to the public, and have a grand wedding on the set; but instead traps Shanti on the set and sets it on fire to prevent a financial loss and to protect his career. Shanti tearfully begs Mukesh to release her but he leaves anyway.

A 12-year-old street kid named Municipality, while on the run from the reformatory, finds and saves a two-day-old abandoned child from being prey to a ferocious street dog. Failing to find any takers among the people he deemed responsible & respectable, Municipality takes up the onus of finding the child's mother himself. The film is an intimate look into the life and manners of the Parsi community especially those living in the city of Bombay (now Mumbai) in the 1950s and 60s. Phirojshah (Naseeruddin Shah) and Pestonjee (Anupam Kher) are close friends. They like to do everything together (and even plan to get married on the same day), even though they are very different in temperament. Phirojshah (Phiroj) is quiet, thoughtful, and indecisive, while Pestonjee (Pesi) is an extrovert. Phiroj is unable to make up his mind about marrying a girl, Jeroo (Shabana Azmi), selected for him by a matchmaker. It is Pesi who ends up marrying her. Phiroj has fallen in love with her, but does not grudge Pesi his happiness. He decides not to marry, and moves to another city. They keep in touch by mail. Phiroj is delighted to hear that they are expecting a baby. He decides to travel to Bombay to visit them. During his visit, Phiroj learns that the married life of Pesi and Jeroo is not as he had imagined it. Jeroo has had a miscarriage. Phiroj is horrified to discover that Pesi has a mistress. He is saddened by how Jeroo has changed. She is no longer the pretty girl she was, but appears cantankerous and uncaring. Both she and Pesi are unhappy in their marriage. Phiroj resolves not to meet them, but mentally is unable to keep himself aloof. All three carry on with their unhappy lives until Pesi's sudden death. Finally, Phiroj discovers that he cannot help Jeroo. Soona, Pesi's mistress whom Phirojshah had thought of as a loose woman, turns out to be the one person capable of genuine love.

Karan (Saif Ali Khan) is a cartoonist and a self-styled ladies' man. His daily comic, named "Hum Tum," explores the battle of stereotypical male/female behavior. On a plane from Delhi to New York, he meets Rhea (Rani Mukerji) who doesn't seem to be interested in him. His feeble attempts at flirting go nowhere, but when they have a stop-over in Amsterdam, she agrees to explore the city with him. Karan quickly learns he has little in common with the feisty but proper Rhea, but he won't give up. He ends their contentious time together with an unwelcome kiss. Outraged, Rhea slaps him, and storms off, but Karan insists they'll meet again. After a few months he spots her in a park in New York, and she makes a scene with his girlfriend (Shenaz Treasurywala), who turns out to be Rhea's childhood friend, which ends in their break-up.

Saikumar plays the role of Prathapa Verma, a rich man in Dubai who has a son Manu (Bala) but also has an adulterous relation with a lady in Mumbai, Janet, with whom he has another son named Alexander (Mohanlal). In his will, he has bequeathed all his wealth to his son Alexander. If Alexander dies, then 40% of Prathapa Verma's wealth will go to his relatives (Siddique, K. B. Ganesh Kumar, etc.) and 30% goes to Manu, and 30% to a trust. If Manu dies, then 100% will go to the trust. So the relatives decide to kill him, while Manu decides to go and meet Alexander and power of attorney from him so that he can get all Verma's wealth, and then kill him or get rid of Alexander. But to their surprise, Alexander is in mental rehabilitation center under the treatment of Dr. Korah (Nedumudi Venu). Eventually, over time, Manu realizes that his brother has unique abilities, which he grows to accept. He starts becoming fond of his half-brother. The story then progresses to show how Manu brings Alexander to Dubai. After reaching Dubai, the struggles that Alexander faces form the rest of the story.
