The film was created by re-dubbing footage filmed for the 1986 Taiwanese kung fu film Ren zhe da (known in English as Ninja: The Final Duel).
Ren zhe da producer Robert Tai had reportedly filmed 10 hours of footage for the film that was cut to 90 minutes for release. Much of the footage that was not included in the release of Ren zhe da is included in Shaolin Dolemite as well as original footage of Moore observing the story of the film and making obscenity-laced comments. Other new footage includes a segment portraying Moore's long-time friend Jimmy Lynch as a Drunken Master named Sam the Spliff.
The re-dubbed plotline involves Tupac (Eugene Thomas), a renegade member of the Dolemite clan, stealing a sacred bell from the Wu-Tang clan and joining forces with a group of ninjas (including the topless Ninja Ho) to defeat a Shaolin temple, which is defended by a Japanese Prince Sanada (Alexander Lou), two white members of the Shabazz clan (Toby Russell and John Ladalski), a coonskin cap wearing man named Davey Crockett (Silvio Azolini), as well as its own monks.

Two sewage workers, Jack Do (Alexander Lou) and Charlie Wu (Charlema Hsu) get mixed up in heavy business when their mobster boss gets assassinated by another mobster who wants to control Shanghai and overflow the streets with opium, along with the usual mafia-related affairs like gambling and prostitution. Advertising company staff Mo Tak-fai and his friend Lai Chi are going to mainland China for a business trip during which Chi smuggled some relics which leads them being chased by the police and they strayed into a cave. Chi was bitten by a poisonous snake in the cave but was saved by medicine from hero Lung Kim-fei from centuries ago and shares it with Tak-fai. Not only does the medicine detoxifies Chi, it also gave Tak-fai and Chi internal strengths. There, they also rescue Princess Wan-lo and her maid Siu-man who were frozen for eight hundred years from the Yuan Dynasty while also allowing two-hundred-year-old evil martial arts expert Tin-chan escape.

In 1881, Chon Wang (Chan) is a Chinese Imperial Guard in the Forbidden City. After Princess Pei-Pei, for whom Wang has affection, is abducted and taken to the United States, the Emperor of China sends three of his guards to retrieve her. Wang is not one of the three, but he tells the Captain of the Imperial Guards that it was his fault the princess was kidnapped and insists on joining them. The Captain first refuses, but when the Royal Interpreter, Wang's uncle, offers to allow Wang to come help with the baggage, the Captain agrees in the hopes that the "foreign devils" would get rid of Wang. In Nevada, Roy O'Bannon (Wilson) is an outlaw who, with his gang, hijacks the train Wang is on. When Wallace, a member of Roy's gang, kills Wang's uncle, Wang chases the outlaws down. However, the gang is well-armed and Wang's only choice is to unhinge the cars and get away on the engine. In the process, Wallace takes over the gang from Roy, and they leave him buried up to his chin in the desert sand. Meanwhile, Pei-Pei, who was tricked into believing she was freely escaping her arranged marriage in China, finds out she has been kidnapped by an agent of Lo Fong, who ran away from the Forbidden City and was viewed as a traitor by the Chinese.

Bu (Shu Qi) is a beautiful young girl from a small Taiwanese fishing village who discovers a romantic message in a bottle. Hong Kong treasure hunter Jackie, a.k.a. "Asian Condor", is summoned by Baron Bannon at his mansion in Madrid, Spain, where he is told of a story of a German commander named Hans von Katterling and his regiment burying 240 tons of gold at a secret base deep in the Sahara Desert in Africa before the end of World War II. The 18 soldiers involved in the operation disappeared under mysterious circumstances. By request from the United Nations, Bannon gives Jackie an unofficial mission to locate the base and recover the gold. Aside from acquiring the key to the base, he is partnered with Ada, an expert in African geography. Upon discovery of the gold, Jackie is promised one percent of the treasure, or roughly 2.5 tons of gold.
