After being falsely accused of his brother Harry's murder Carl Pearson escapes from prison with the aid of another prisoner Duke Williams. Meanwhile wealthy aircraft manufacturer Roger Morgan makes plans to adopt Harry's son Bobby who is currently in the care of Ruth Adams, who is also Carl's fiancee. With his father dead young Bobby is the legal owner of valuable aircraft plans that Morgan wants to acquire. Carl's son is Johnny who is the owner of Rinty a prize tracking dog. The Police borrow Rinty in order to try to track Carl and Duke; but Rinty is less than cooperative in this effort. After a car accident leaves Ruth temporarily incapacitated, young Bobby wanders off. A canoe rescue and an encounter with a wild cougar follow. Arriving at the Pearson's cabin Carl recovers the plans, but Duke steals them in order to sell them. Eventually everyone else ends up at the cabin and the real killer is revealed. The killer tries to escape followed by Carl and Rinty in a climactic chase.

In this serial, the intrepid Buffalo Bill Cody comes to aid miner Rocky Ford and a group of ranchers in their defeat of a local crime lord, King Carney, who is trying to keep the new railroad out of the territory in order to carry on with his illegal operations. Roar of the Iron Horse had hero Jock Mahoney in his first leading role, playing Jim Grant, a railroad agent, opposing a typical hardfaced German, Karl Ulrich, called The Baron. Head of a strong ring in America, the infamous Baron was thwarted time and time again as he tried to sabotage the building of the transcontinental railroad with all the means to his scope, strategically bribing the local Indians into doing his dirty work. The Bascom Ranch, owned by Texas Rose Bascom, is targeted by an outlaw gang with the intent to steal cattle off the ranch. Outlaw Freno Frost runs the rustlig gang. One of the gang members is Jim Bascom, Texas Rose Bascom's wayward brother. When she discovers that her brother is running with the outlaws, she seeks help from the law.
