Anthony "Tick" Belrose (Hugo Weaving), using the drag pseudonym of Mitzi Del Bra, is a Sydney-based drag queen who accepts an offer to perform his drag act at Lasseter's Hotel Casino Resort managed by a female friend named Marion in Alice Springs, a remote town in central Australia. After persuading his friends and fellow performers, Bernadette Bassinger (Terence Stamp), a recently bereaved transsexual woman, and Adam Whitely (Guy Pearce), a flamboyant and obnoxious younger drag queen who goes under the drag name Felicia Jollygoodfellow, to join him, the three set out in a large tour bus, which Adam christens "Priscilla, Queen of the Desert" for a four-week run in the Australian Outback town.

In the 1930s, Teresa (Buday) is a naive young woman dealing with the oppressive attitudes of society and her father's austere ways. Louis Salinger, of Interpol, and Eleanor Whitman, an Assistant District Attorney from Manhattan, are investigating the International Bank of Business and Credit (IBBC), which funds activities such as money laundering, terrorism, arms trading, and the destabilization of governments. Salinger's and Whitman's investigation takes them from Berlin to Milan, where the IBBC assassinates Umberto Calvini, an arms manufacturer who is an Italian prime ministerial candidate. The bank's assassin diverts suspicion to a local assassin with political connections, who is promptly killed by a corrupt policeman. Salinger and Whitman get a lead on the second assassin, but the corrupt policeman shows up again and orders them out of the country. At the airport they are able to check the security camera footage for clues on the whereabouts on the bank's assassin, and follow a suspect to New York.

He then awakens in the Australian outback and sees a white mare. He rides it into town, only to be arrested and subsequently imprisoned in 1871, for supposedly stealing the horse, even though it had actually been stolen by Wild Wright, Ned's friend.
