Two American college students, David Kessler (David Naughton) and Jack Goodman (Griffin Dunne), backpack across the North York Moors. As darkness falls, they stop for the night at a pub called "The Slaughtered Lamb". Jack notices a five-pointed star on the wall. When he asks about it, the pubgoers stop talking and become hostile, telling them to keep to the road and beware of the full moon. The pair decide to leave, but while talking they wander off the road onto the moors.

In 1895, a young immigrant couple is refused entry into Manhattan because they have consumption. Charles Brady and his mother Mary are Sleepwalkers—nomadic, shapeshifting energy vampires who feed off the lifeforce of virgin women. Though they normally maintain a human form, they can transform (partially or fully) into human-sized bipedal werecats (presumably their natural form) at will. They are considerably more resilient than humans and have powers of both telekinesis and illusion. Their one weakness is cats, who are not only able to see through their illusions but whose claws are capable of inflicting severe to fatal wounds upon them. It is revealed that they also maintain an incestuous relationship.

While playing with Copper at his home, Tod awakens Chief. Slade and Chief chase him until they are confronted by Tweed. After a violent argument, Slade threatens to kill Tod if he trespasses on his farm again. Hunting season comes and Slade takes his dogs into the wilderness for the interim. Meanwhile, Big Mama, Dinky and Boomer attempt to explain to Tod that his friendship with Copper can no longer continue, as they are natural enemies, but Tod naively refuses to believe them, hoping that he and Copper will remain friends forever.

Kham is the last of a family line of guards who once watched over the King of Thailand's war elephants. In 1939 New Orleans, a roguish German Shepherd named Charlie B. Barkin escapes from a dog pound with the help of his friend, a dachshund named Itchy Itchiford. They return to a casino riverboat on the bayou, which is formerly run by Charlie with his partner, a bulldog named Carface Caruthers. Not wanting to share the profits with Charlie, Carface persuades him to leave town with 50% of the casino's earnings. Charlie agrees, but is later intoxicated and murdered by Carface. He is sent to Heaven where a whippet angel named Annabelle (who was named in the sequel) tells him that a gold watch representing his life has stopped. He steals and winds it, sending Charlie back to Earth, but is told that if he dies again, he will go to Hell. After reuniting with Itchy, they discover that Carface is holding an orphan girl named Anne-Marie (presumably kidnapped from the orphanage by Carface), who has the ability to talk to animals and gain knowledge of a race's results beforehand, allowing Carface to rig the odds on the rat races and become rich. They rescue her, intending to use her abilities to get revenge on Carface, though Charlie tells her that they plan to give their winnings to the poor and help her find parents.

In the French Alps, during the year 1943, Sébastien is a seven year old orphan boy living with César, an adoptive "grandfather" and his daughter, Angélina. One day, on the way home, he meets a huge dog, wild and completely discredited: it is the so-called "Beast". Sébastien is not slow to make friends with the animal, a female sheepdog, and names her "Belle" because he is struck by her beauty after she is cleaned up from the mud that she was coated with. Sebastian decides to keep his friendship with Belle secret to protect her.

Andy McDermott is a tourist seeing the sights of Paris with his friends Brad and Chris. When Serafine Pigot leaps off the Eiffel Tower just before Andy is about to bungee jump, he executes a mid-air rescue. She vanishes into the night, leaving Andy intrigued – and also unaware that she is the daughter of David Kessler and Alex Price, the couple seen 16 years earlier in the first film. That night, Andy, Chris, and Brad attend a night club called "Club de la Lune". The club's owner, Claude, is actually the leader of a werewolf society that uses the club as a way to lure people (preferring tourists) in to be killed. Serafine arrives, tells Andy to run away and transforms into a werewolf. The club owners transform into werewolves as well and butcher all the guests. Chris escapes and goes back to Serafine's house. Brad's heart is eaten by a werewolf and Andy is bitten by another werewolf.
