Adam (hébreu אָדָם, araméen/syriaque ܐܕܡ, arabe آدم) est un personnage du Livre de la Genèse et du Coran. Dans ces textes, qui fondent la mythologie biblique et les croyances juives, chrétiennes et musulmanes, il est le premier homme, créé par Dieu. Il meurt à 930 ans (Genèse : 5-5), à 960 ans selon un hadith. Il est également le compagnon d'Ève, créée à partir de son corps.

Jacob (Doug Bradley) is an antiques dealer who sets off with his new young and pregnant wife Lauren (Grace Vallorani) and his stepdaughter, Rachel (Rita Ramnani) to a forest homestead in a hopeful effort to heal bonds between feuding family members. He also carries a large, mysterious and ancient Babylonian mirror, smuggled out of the middle east, which he plans to sell for profit. Lilith (Natalie Celino), a sultry ornithologist appeares out of a bird hide by a noisy pair of friends hiking through the woods. Gaining their confidence, she and one of the men, Travis (Scott North) go into the woods for a quick tryst, but immediately there is a blood curdling scream. The other camper, Stanley (James Fisher), rushes toward the sound and finds his friend mutilated and near dead. Near Travis, Lilith explains to Stanley that the shadows had come to life and attacked them, and the two set off to seek help. When they end up at Jacob's cabin, the entire group comes under attack by supernatural forces of darkness. Rachel discovers Irish cowboy Phelan (Jonnie Hurn) hanging out in their barn. Phelan is a vampire and himself a former vampire hunter. He tells Rachel that the real danger to everyone is Lilith, and explains how Lilith had been Adam's willful first wife who, since having been exiled from the Garden of Eden, had been spawning demonic offspring in the world of man. Considered the mother to the Succubi, ruler of shadows, slaughterer of children, and consort to the devil himself, she had been re-awakened by, and drawn to the location of, the occult power inherent in the mirror carried by Jacob. Phelan has been harboring a centuries-long grudge against her for she having "turned" him 120 years earlier, and he had been tracking her for decades, equipped with the one relic that might finally kill her... a bone splinter from the rib of Adam himself.

Zed (Jack Black) is a hunter and Oh (Michael Cera) is a gatherer. After being informed that Zed ate from the Tree of Knowledge of Good and Evil, the shaman (Bill Hader) and Marlak (Matthew Willig) banish him from the tribe. After Zed burns down the village by accident, Oh decides to go with Zed on his journey to discover all the world has to offer. Along the way, they encounter Cain and Abel (David Cross and Paul Rudd). Cain kills Abel and informs Zed and Oh that they must escape with him or else be accused of killing Abel.

In the Beginning is the story about the travels and travails of the tribe of Abraham (Martin Landau). Set around the year 2000 B.C., the narrative opens with "Genesis 12," wherein the LORD has told Abraham and company to leave their country to a land of milk and honey to be named later. In order to keep up the morale of his followers while on the road, Abraham gives a sermon that sums up God's creation of the universe. By illustrating this sermon with stock footage and special-effects shots, the producers attempt to make a connection between sermons of old and popular entertainments of today. From there, the twists and turns of the Old Testament are treated like a soap opera. Family dramas take center stage, whether it's God testing Abraham by telling him to kill his son in sacrifice, Joseph gaining power in Egypt after being sold to slave traders by his brothers, or one of the many other stories of brothers fighting (Cain and Abel, Isaac and Ishmael, etc.). Many events are visualized such as the plagues and the parting of the Red Sea, to name but two.

The film is a "spoof of the creation of life". Adam is a man who is created by the Hand of God and is put on this lonely planet. He does silly acts and does all sorts of strange things. When God realizes that Adam is lonely, he makes Adam a friend. Adam, expecting it to be Eve, gets himself ready as if for a date only to find out his new friend is a penguin.

The film consists of five main sections: The Creation, Adam and Eve, Cain and Abel, Noah's Ark, and the story of Abraham. There are also a pair of shorter sections, one recounting the building of the Tower of Babel, and the other the destruction of Sodom and Gomorrah. The sections vary greatly in tone. The story of Abraham is somber and reverential, while that of Noah repeatedly focuses on his love of all animals—herbivorous and carnivorous or omnivorous. Cats (including lions) drink milk, with Noah's relationship with the animals being depicted harmoniously. It was originally conceived as the first in a series of films retelling the entire Old Testament, but these sequels were never made.
