In Venice at the beginning of the 17th century, the Three Musketeers, Athos (Matthew Macfadyen), Porthos (Ray Stevenson), and Aramis (Luke Evans), with the help of Athos' longtime lover, Milady de Winter (Milla Jovovich), steal airship blueprints made by Leonardo da Vinci. However, they are betrayed by Milady, who gives the blueprints to the Duke of Buckingham (Orlando Bloom). Upon returning to France, the Musketeers are forced to disband by Cardinal Richelieu (Christoph Waltz) for their failure, and they end up on the streets of Paris.

As a young boy, d'Artagnan witnesses the murder of his parents at the hands of Febre (Tim Roth), chief henchman of Cardinal Richelieu (Stephen Rea). d'Artagnan is nearly killed after using his dead father's sword to fight Febre which leaves him with a permanent scar and blinds him in one eye. d'Artagnan is taken in by family friend Planchet (Jean-Pierre Castaldi), a former musketeer, one of the loyal protectors of King Louis XIII (Daniel Mesguich).

In the autumn of 1654, an escaped slave seeks refuge at a convent in southern France. After the nuns take him in, his pursuers break into the convent and assault the nuns, searching for the slave. After wounding the mother superior (Pascale Roberts), they close in on their prey. One feisty novitiate, Eloise (Sophie Marceau)—the daughter of the renowned swordsman D'Artagnan (Philippe Noiret) of the famed Three Musketeers—stands up to the intruders, but is shoved aside as they ride off after the frightened slave, who escaped from the evil Duke Crassac de Merindol. Eloise finds a blood-stained piece of paper (a simple laundry list) that the slave used to stop a bleeding wound, and believes it holds some secret code. Later, Eloise is with the mother superior when she dies from her wound. Swearing a sacred oath to avenge her and dressed in men's clothes, Eloise sets out for Paris to elicit the help of her father in tracking down the murderers.

During a war between France and the Protestant rebels of La Rochelle, Cardinal Richelieu orders Count de Rochefort to kidnap Constance Bonancieux. The evil Milady de Winter, who wants revenge on d'Artagnan, seduces him to keep him occupied. The young d'Artagnan arrives in Paris with dreams of becoming a king's musketeer. Quite unused to the city life, he makes a number of clumsy faux-pas. First he finds himself insulted, knocked out and robbed by the Comte de Rochefort, an agent of Cardinal Richelieu, and once in Paris comes into conflict with three musketeers, Athos, Porthos, and Aramis, each of whom challenges him to a duel for some accidental insult or embarrassment. As the first of these duels is about to begin, Jussac arrives with five additional swordsmen of Cardinal Richelieu's guards. D'Artagnan sides with the musketeers in the ensuing street fight and becomes their ally in opposition to the Cardinal, who wishes to increase his already considerable power over the king. D'Artagnan also begins an affair with his landlord's wife, Constance Bonacieux, who is the Queen's dressmaker.

Première époque — Les Ferrets de la reine Au XVII siècle, le jeune D’Artagnan quitte sa Gascogne natale pour Paris où il va postuler auprès du capitaine des mousquetaires, M. de Tréville, une connaissance de son père. Sur son chemin, il a une altercation avec un mystérieux individu qu’il a aperçu s’entretenant avec une non moins mystérieuse jeune dame blonde et on lui dérobe la lettre de recommandation destinée à M. de Tréville. Engageant Planchet comme valet pour continuer sa route, d'Artagnan arrive à Paris, vend son cheval boiteux et se rend à la caserne des mousquetaires. Pendant l'entretien avec le capitaine, il reconnaît son agresseur et le poursuit pour se venger. Mais dans sa course, il s'achoppe successivement à trois mousquetaires, Athos, Porthos et Aramis. Comme l’exige la coutume des mousquetaires, bien que les duels soient interdits par le cardinal de Richelieu, ceux-ci le défient pour réparation. Il accepte avec grâce, devant se battre à midi, une heure et deux heures avec chacun d'entre eux au Pré-aux-Clercs, derrière le jardin du Luxembourg. Sur les lieux de leurs affrontements, les quatre hommes sont surpris par les gardes du cardinal et pactisent alors pour les combattre. Vainqueurs, les trois mousquetaires et d’Artagnan deviennent amis, s’étant reconnus comme de fidèles sujets de leur roi Louis XIII. Ils vont, dès lors, être les défenseurs acharnés du couple royal et, de ce fait, devenir les ennemis jurés du cardinal et de ses nombreux sbires, dont le comte de Rochefort, le capitaine des gardes du cardinal, et la très dangereuse autant que belle espionne Milady de Winter…

M. de Tréville mécontent que ses mousquetaires Athos, Porthos et Aramis se soient encore battu avec les hommes du cardinal, les convoque pour les sermonner. Le jeune D’Artagnan profite de la situation pour demander d’être sous les ordres de M. de Tréville mais il lui annonce qu'on lui a dérobé sa lettre de recommandation. En effet, montant un cheval jaune ou bouton d'or, il a provoqué le comte de Rochefort en duel à Meung, qui a préféré lui donner le bâton par ses valets. Pressé, le comte s’entretenait avec Milady de Winter. Sortant de son entrevue avec M. de Tréville, D’Artagnan bouscule et se chamaille avec chacun des trois mousquetaires, ce qui lui vaut un duel avec chacun d'eux au Luxembourg. Sur le lieu de leur affrontement, les quatre hommes sont surpris par les gardes du cardinal et pactisent alors pour les combattre sous le cri de « Tous pour un, un pour tous ». Les gardes sont accompagnés de Lord de Winter, le beau-frère de Milady, qui combat à leur côté contre D’Artagnan. Mais finalement, les deux hommes fraternisent et le Lord invite le Gascon chez sa sœur qui tombe sous son charme.

Young d'Artagnan leaves his parents and travels from his native Gascony to the capital of France because he wants to prove himself an excellent fencer and to become a musketeer. He is told by his father he must not avoid any duel. On his way to Paris, d'Artagnan feels that his honour is besmirched because he overhears how his horse is derided by a sinister nobleman. He can't help but demand immediate satisfaction. Unfortunately, of all men he finds he has challenged the Count de Rochefort, a shifty character to whom Cardinal Richelieu frequently entrusts covert operations. Rochefort's henchmen take care of d'Artagnan and steal from him. The enraged d'Artagnan is determined to take revenge and will eventually have the chance to do so, for the Queen has given a present to her secret admirer the Duke of Buckingham, and d'Artagnan must retrieve it from him, although he is now already back in England. If he fails her, Cardinal Richelieu is going to disclose Queen Anne's infidelity to King Louis XIII, in order to force a war against England upon him. The Cardinal and Count de Rochefort will do everything in their power if only they can put paid to d'Artagnan's mission. But with help from his three new friends d'Artagnan prevails.
