Five years after the first film, Lou Dorchen, Nick Webber, and Jacob have led their lives to fame, but each struggling with their self-esteem. Lou brags his success through time travel to his employee Brad, while Nick's wife Courtney complains about Nick going wherever he wants, whereas Jacob shows hatred for Lou. Lou hosts a party where he declines an offer by old schoolmate Gary Winkle to buy land, and forcing Jacob to act as a butler. However, Lou is shot in the penis, urging Jacob and Nick to take him to the hot tub time machine, in hopes to find and stop Lou's killer.

The film opens with Abby Russell at a night club where she explains to the audience via a narration that her "job" is to murder cheating men in order to "keep scum off of the streets"- a job that she thoroughly enjoys. She then murders a man by slicing through his femoral artery and then throwing him off the roof of the club. The following day she attends a ceremony where she watches Danni Rodgers, a nurse she had been mentoring, graduate. She then approaches her and meets Danni's mother and stepfather Larry Cook. Despite this joyful moment and all of her preparation, Danni is ill prepared for her first true day of nursing duty and is berated by Dr. Morris for her inability to respond quickly to an emergency situation. Abby views the situation with disgust, as she knows that Dr. Morris is a sadist that enjoys harassing new nurses.

In order to enjoy his retirement, a father takes drastic measures to get his twenty-something, slacker sons to move out and fend for themselves. They continue to try and make their own film company, called Cathkart productions. Tom Cathkart (Danny DeVito) eventually gets sick of his sons slacking off and he leaves them alone and takes all the food with him, leaving the sons to no option get a job, they decide to open their own bed and breakfast and they let their friends stay at the house for money. Eliot (Ryan Hansen) falls in love with Sarah (Caitlin Crosby) and his brother Quinn (Skyler Stone) gets jealous of his success with the film company. Eliot then leaves and his brother finds it hard to manage without him.

Following the events of Harold & Kumar Go to White Castle, Harold Lee and Kumar Patel set off on a flight to Amsterdam so that Harold can pursue a budding romance with his neighbor, Maria. Before they board their plane, they run into a friend from college named Colton, and Kumar's ex-girlfriend, Vanessa, who are getting married in Texas. During the flight, a woman suspects Kumar is a terrorist after mistaking his bong for a bomb. Sky marshals detain the duo and the plane diverts to Washington D.C. Ron Fox, an obsessive and racist Deputy Secretary of Homeland Security, sends them to Guantanamo Bay detention camp in Cuba, thinking they are agents of a joint Al-Qaeda and North Korean conspiracy, against the advice of NSA Vice-Chairman John Beecher. With the inadvertent assistance of two other prisoners, Harold and Kumar escape and board a Cuban refugee boat to Miami, Florida. They meet up with their college friend, Raza, who loans them a car so they can travel to Texas to get help from Colton, whose family has political connections.

Dewey Cox begins his quest for stardom in Springberry, Alabama in 1946. While playing with his brother Nate, Dewey accidentally cuts his brother in half at the waist with a machete. Bartleby Gaines (Justin Long) is a persuasive senior from William McKinley High School in Wickliffe, Ohio, who, among other pranks, creates fake IDs. His gifts do not extend to grades, however, and he receives rejection letters from all of the colleges to which he applies, including those with high acceptance rates. In an attempt to gain approval from his strict father (Mark Derwin), Bartleby creates a fake college, the South Harmon Institute of Technology (S.H.I.T.). His best friend, Sherman Schrader III (Jonah Hill), who has been accepted into his father's prestigious alma mater Harmon College, aids Bartleby and fellow rejects Rory Thayer (Maria Thayer), who only applied to Yale University; Hands Holloway (Columbus Short), who lost his athletic scholarship after an injury; and Glen (Adam Herschman), who failed his SAT exam. To make the "college" seem legitimate, Bartleby convinces Sherman to create a functional website for the school.
