Two years after their escapade in Las Vegas, Stu Price, Phil Wenneck, Alan Garner and Doug Billings travel to Thailand to celebrate Stu's upcoming wedding to Lauren. Much to Alan's dismay, they are joined by Lauren's younger brother, Teddy. Lauren's father's reveals his disapproval of Stu during a toast. At the end of the night, Stu hesitantly joins Phil, Doug, Alan and Teddy for a beer. Sitting at a campfire and roasting marshmallows, the group toast to Stu and Lauren's future happiness.

Andy, now 17 years old, is about to leave for college, and his toys have not been played with for years. He intends to take only Woody with him, and puts Buzz Lightyear, Jessie and the other toys in a bag to be stored in the attic. Andy's mother mistakenly takes the bag to the curb for garbage pickup. The toys escape and, believing Andy intended to throw them away, decide to climb in a donation box with Barbie bound for Sunnyside Daycare. Woody follows them and tries to explain the mistake, but they refuse to believe him.

A dust speck is dislodged from its obscure place and sent adrift through the Jungle of Nool. At the same time, Horton the elephant (Jim Carrey), the jungle's eccentric nature teacher, takes a dip in the pool. The dust speck floats past him in the air, and he hears a tiny yelp coming from it. Believing that an entire society of microscopic creatures are living on that speck, he places it atop a clover. Horton finds out the speck harbors the city of Whoville and its inhabitants, the Whos, led by Mayor Ned McDodd (Steve Carell). He has a wife, Sally (Amy Poehler), 96 daughters (whose names all begin with the letter H), and one teenage son named JoJo (Jesse McCartney). Despite being the oldest and next in line for the mayoral position, JoJo doesn't want to be mayor, and because he's so scared of disappointing his father, never speaks. Once Horton begins carrying the speck with him, the city starts experiencing strange phenomena (earthquakes and changes in the weather), and the Mayor finds his attempts to caution Whoville challenged by the Town Council, led by the opportunistic yet condescending Chairman (Dan Fogler).

A documentary crew follows the events of Cody Maverick, a 17-year-old rockhopper penguin who lives in Shiverpool, Antartica with his mother Edna and his older brother Glen. Cody has wanted to be a professional surfer ever since a visit from legendary surfer Ezekiel "Big Z" Topanga several years ago. When a talent scout shorebird named Mikey (Mario Cantone) arrives to find entrants for the "Big Z Memorial" surfing contest, Cody jumps at the chance despite the lackluster support from his family. Winnie the Pooh and his friends hear a strange noise and find a set of large, circular footprints in the Hundred Acre Wood. During the night, Tigger's house is damaged by what appears to be an earthquake. The friends believe that there is a heffalump in the woods. Rabbit organizes an expedition to go try to catch it. Roo wants to comes long, but the others tell him he is too young and small to go. Despite this, Roo slips out on his own in search of the heffalump. He finds one; a playful young creature named Heffridge Trumpler Brompet Heffalump IV - "Lumpy" for short. Roo is afraid of his captive at first, but the two quickly become friends and play.

Maggie is the only cow left on the Dixon Ranch after Alameda Slim (a cattle rustler capable of stealing 500 in a single night) stole all the rest of Mr. Dixon's cattle. Dixon sells Maggie to Pearl, a kind and elderly woman who runs a small farm called Patch of Heaven. The local Sheriff arrives to tell Pearl that her bank is cracking down on debtors. Pearl has three days to pay the bank $750, or her farm will be sold to the highest bidder. Hearing this, Maggie convinces the other cows on the farm (Grace, a happy-go-lucky character, and Mrs. Caloway, who has had leadership gone to her head) to go to town to attempt winning prize money at a fair. While the cows are in town, a bounty hunter named Rico (who Buck, the Sheriff's horse, idolizes) drops a criminal off and collects the reward. Stating he needs a replacement horse to go after Alameda Slim while his own horse rests, he takes Buck. When Maggie find out that the reward for capturing Slim is of exactly $750, she convinces the other cows to try to capture him to save Patch of Heaven.
