"Smiler" Grogan (Jimmy Durante), wanted by police in a tuna factory robbery fifteen years ago and on the run from the police, careens his 1957 Ford Victoria off twisting, mountainous State Highway 74 near Palm Springs in Southern California and crashes. Five motorists stop to help him - Melville Crump (Sid Caesar), a dentist, Lennie Pike (Jonathan Winters), a furniture mover, Dingy Bell (Mickey Rooney) and Benjy Benjamin (Buddy Hackett), two friends on their way to Las Vegas, and J. Russell Finch (Milton Berle), who owns Pacific Edible Seaweed Company in Fresno. Just before he dies, Grogan tells the five about $350,000 buried in Santa Rosita State Park near the Mexican border under a mysterious "big W".

The film is a drama set in World War II, following a group of United States Marines from boot camp to combat (6th Marines, 2d Marine Division). It depicts the forming of comradeships and the relentless training which the battalion, led by Major "High Pockets" Huxley, has to undergo. In two battles (Guadalcanal and Tarawa), the battalion is used only for mop-up duties, but after an out-of-line argument between "High Pockets" and the commanding general, they are used as lead battalion in the battle for Saipan, suffering heavy casualties as they land on the exposed left flank of Red Beach. Huxley is killed and as they watch his body being carried off the field his grim-faced men fix bayonets ready to meet a Japanese counter-attack.

Esther Blodgett is a talented aspiring singer with a band, and Norman Maine is a former matinee idol with a career in the early stages of decline. When he arrives intoxicated at a function at the Shrine Auditorium, the studio publicist attempts to keep him away from reporters. After an angry exchange, Norman rushes away and bursts onto a stage where an orchestra is performing. Blodgett takes him by the hand and pretends he is part of the act, thereby turning a potentially embarrassing and disruptive moment into an opportunity for the audience to greet Norman with applause.

Sach is punched in the nose by Herbie, a local kid who the boys know, and acquires the ability to read people's minds. Slip sees this as an opportunity to buy a detective agency and their first client is a beautiful blonde who is trying to escape from her connections with mobsters. She leaves behind a stolen mink coat and an envelope that would incriminate the mobsters. The mobsters, trying to get the envelope back, kidnap Herbie in the hopes to persuade the boys to return the envelope. Slip and Sach, after being tipped off where Herbie is being held, go there in disguise. They foil the mobsters plans and rescue Herbie.

The basic plot remains the same as in the stage play and the 1936 film version. When the Cotton Blossom, Cap'n Andy Hawks's show boat, arrives in a Mississippi town to give a performance, a fistfight breaks out between leading man Steve Baker (Robert Sterling), and Pete (Leif Erickson), the boat's engineer who has been making passes at Steve's wife, leading lady Julie La Verne (Ava Gardner). But Cap'n Andy (Joe E. Brown) pretends to the assembled crowd that the two were really previewing a scene from one of the boat's melodramas. But Pete knows a dark secret about Julie, and he runs off to tell the local sheriff.
