Eric (Jason Sudeikis) is a perpetual adolescent who lives to party, holding lavish theme-events with his friends using his father's large house in the Hamptons. On July 3, Eric throws a white trash bash party attended by his friends Sue (Michelle Borth), Adam (Nick Kroll), Mike (Tyler Labine), Laura (Lindsay Sloane), Kate (Lucy Punch), Glenn (Will Forte), Doug (Martin Starr) and his girlfriend Willow (Angela Sarafyan), Alison (Lake Bell) and her boyfriend Marcus (Rhys Coiro). Eric meets Kelly (Leslie Bibb) at the party. The following morning, Eric's father (Don Johnson) arrives and informs him that he is selling the house. The next weekend, the group arrive back at the house and find it for sale by Dody (Lin Shaye) and Kelly.

Under the watchful eyes of Father Silvano Bentivoglio (Carmen Argenziano) and Dr. Vittoria Vetra (Ayelet Zurer), the European Organization for Nuclear Research (CERN) initiates the Large Hadron Collider and creates three vials of antimatter particles larger than any that have ever been produced before. Almost immediately, Father Silvano is killed and one of the vials of antimatter goes missing. At the same time, the Roman Catholic Church is mourning the sudden death of Pope Pius XVI in Rome and prepares for the papal conclave to elect the next Pope. The Camerlengo Patrick McKenna (Ewan McGregor) assumes temporary control of the Vatican. The Illuminati kidnap four of the 'preferiti' (the favourite cardinals to be elected pope) before the conclave enters seclusion and threaten to kill one candidate every hour and destroy all of Vatican City at midnight, using the missing vial of antimatter as a bomb. The Vatican summons symbologist Robert Langdon (Tom Hanks) from Harvard University and Vittoria from CERN to help them save the four preferiti and locate the vial.

In Los Angeles in 1928, single mother Christine Collins (Jolie) returns home to discover her nine-year-old son, Walter (Gattlin Griffith), is missing. Reverend Gustav Briegleb (Malkovich) publicizes Christine's plight and rails against the Los Angeles Police Department (LAPD) for its incompetence, corruption and the extrajudicial punishment meted out by its "Gun Squad" led by Chief James E. Davis (Feore). Several months after Walter's disappearance, the LAPD tells Christine that he has been found alive. Believing the positive publicity will negate recent criticism of the department, the LAPD organizes a public reunion. Although "Walter" (Devon Conti) claims he is Christine's son, she says he is not. Captain J. J. Jones (Donovan), the head of the LAPD's Juvenile Division, insists the boy is Walter and pressures Christine into taking him home "on a trial basis".
