Walter Mitty is a negative assets manager at Life magazine who daydreams of adventures and has a crush on a coworker named Cheryl. Mitty works with photojournalist Sean O'Connell, whose images are highly regarded. O'Connell has sent Mitty his latest negatives and a wallet as a gift in appreciation of Mitty's work. O'Connell believes negative #25 captures the "quintessence" of Life and that it should be used for the cover of the magazine's final print issue as it converts to online status. The negative is missing, however, and Walter is forced to stall for time with corporate transition manager Ted Hendricks, who is handling the downsizing. While viewing the other negatives outside Life's offices, Cheryl approaches Mitty and suggests that he think of the negatives as clues to Sean's location. They look at three of them, including one of a person's thumb with a unique ring on it, and another of a curved piece of wood. A third picture of a boat leads Mitty to determine that O'Connell is in Greenland. Mitty promptly flies there to find him.

Andy Brewster (Seth Rogen) is a UCLA-graduate organic chemist and inventor. He is attempting to get his environmentally friendly cleaning product, ScioClean, in a major retail store. However, each retail store he visits dismisses him before he can end his pitch. After a disappointing sales pitch to K-Mart, he visits his mother, Joyce Brewster (Barbra Streisand), in New Jersey before leaving on a cross-country trip to Las Vegas, lying to her that his pitch ended well so she won't worry about him. While there she reveals to him that he was named after a boy she fell in love with in Florida named Andrew Margolis, whom she hoped would object to her marriage with Andy's father. However, he never did and she felt that she never mattered to him afterwards. After a little research, he finds Andrew Margolis is still alive and unmarried living in San Francisco. He invites his unknowing mother on the trip, claiming he wants to spend some time with her.

In eastern Siberia's Dzhugdzhur Mountains, Dieter Von Cunth and his men take control of the X-5 missile, which is a nuclear warhead. On the other side of the world Col. Jim Faith and Lt. Dixon Piper are searching for former Green Beret, Navy SEAL and Army Ranger MacGruber in Ecuador. The two men find MacGruber meditating in a chapel, and try to convince him to return to the United States in an effort to retrieve the warhead. Throughout his entire life, Rod Kimble (Andy Samberg) has believed his father, now deceased, was a successful and respectable stuntman working for Evel Knievel. He aspires to follow in his father's footsteps and become a famous stuntman himself. Meanwhile, his stepfather Frank (Ian McShane) fails to respect Rod as a man, often going out of his way to beat him in sparring sessions and mocking his stuntman dream. Rod makes many attempts at landing jumps with his Tomos moped, most of them ending unsuccessfully. After an ill-fated jump attempt at the public pool, he returns home to find out that Frank is in urgent need of a heart transplant. Angry at the thought of his stepfather dying without getting a chance to "smash his face in" and gain his respect, Rod tells his childhood friends Rico (Danny McBride), Dave (Bill Hader) and his half-brother Kevin (Jorma Taccone) about his plan to do a jump over the length of fifteen school buses, and give the proceeds to his stepfather's surgery. After a random attack the night before by a local gang known as "The Scullions" and their infantile leader Zed McGlunk (Bobcat Goldthwait), Chief Henry Hurst (George R. Robertson) arrives at the 16th precinct and notifies its captain, Pete Lassard (Howard Hesseman) that the precinct is the worst in the city. Nevertheless, Hurst gives Lassard 30 days to turn the precinct around or he is out.
