Jerry Maguire (Tom Cruise) is a glossy 35-year-old sports agent working for Sports Management International (SMI). After having a life-altering epiphany about his role as a sports agent, he writes a mission statement about perceived dishonesty in the sports management business and his desire to work with fewer clients so as to produce better quality. In turn, Management decides to send Bob Sugar (Jay Mohr), Jerry's protégé, to fire him. Jerry and Sugar call all of Jerry's clients to try convincing them not to hire the services of the other. Sugar secures most of Jerry's previous clients. Jerry speaks to Arizona Cardinals wide receiver Rod Tidwell (Cuba Gooding, Jr.), one of his clients who is disgruntled with his contract. Rod tests Jerry's resolve through a very long telephone conversation while Sugar is able to convince the rest of Jerry's clients to stick with SMI instead. Leaving the office, Jerry announces that he will start his own agency and asks if anyone is willing to join him, to which only 26-year-old single mother Dorothy Boyd (Renée Zellweger) agrees. Meanwhile, Frank "Cush" Cushman (Jerry O'Connell), a superstar quarterback prospect who expects to be the number one pick in the NFL Draft, also stays with Jerry after he makes a visit to the Cushman home. However, Sugar is able to convince Cushman and his father at the last minute to sign with SMI over Jerry. Cushman's father implies they decided to sign with Sugar over Jerry when they saw Jerry attending to Tidwell; an African-American player, versus his son (a white player).

Twin girls Jessica and Suzy Martin (Mary-Kate and Ashley Olsen) live with their father, Stephen (Patrick Cassidy), in Philadelphia. They have a dream about somebody saying, "This town ain't big enough for the three of us" and having a shootout but argue who makes their move first. One day, they receive a letter from their late mother Sarah's godmother, Natty (Phillips), inviting them to a dude ranch. She, not knowing about Sarah's death, wants her to come help at the ranch. She is having financial problems because the ranch has too few paying visitors and, although she has offers, she does not want to sell it. The girls try to ask their dad's uptight boss, "The Dragon Lady" (Ms. Plaskett), for his vacation time, only to get him fired. They lie and say that she said that they could go. They go to the ranch and attempt to help Natty.

The first portion of the film is set on November 21, 1963 (the day before President John F. Kennedy was assassinated). Birdlace and three of his Marine buddies have arrived in San Francisco for twenty-four hours, before shipping off to Vietnam, and are planning on attending a "dogfight" (a party where Marines compete to bring the ugliest date, unbeknownst to the girls they bring) later that evening. They separate into the city to attempt to find dates. After a few women reject his advances, Birdlace ducks into a coffee shop, where he encounters Rose, a waitress, on her break, practicing her guitar. She is not particularly "ugly", but rather plain, shy and awkward. Birdlace attempts to charm her, complimenting her on her guitar playing, and inviting her to a party. She is suspicious of his motives, but decides to accept his invitation.
