Shirley Moochmoore (Rebecca Gibney) is a sweet misfit and mother of five daughters who are all convinced they suffer from various mental illnesses. The assassination of Tokyo governor Takayama (Mac Yasuda) causes a stir of public outrage in Tokyo, Japan. Upon hearing the news of the incident, the FBI asks the CIA's Tokyo office to investigate the killing, believing it to be linked to the Yakuza, a dangerous Japanese mafia. Retired CIA agent Travis Hunter (Steven Seagal) and rookie FBI agent Sean Mac (Matthew Davis) are assigned to work on the case and to track down the perpetrators. The Japanese branch of the CIA starts sniffing around under the auspices of Homeland Security. During work, Mac proves to be more of a distraction, not really knowledgeable about procedures or Japanese customs. Gary (not his real name, which is never revealed) is a hustler walking through the streets of New York City, looking for business. On the way, he saves a young deaf transvestite from a group of gay bashers, but regrets it afterwards because the transvestite becomes infatuated with Gary and follows him everywhere. Gary introduces the transvestite to his friends: Valentino, a former porn star, and Mary Carmen, the Latina who works as a doughnut shop waitress and is in love with Valentino, with whom she has been living for some time. Together they form a bohemian family, which includes Veronica, a still-active porn star, and Nat, a tattoo artist. Gary is also in love with Valentino, who is dying of AIDS. Through the stages of the disease, Mary Carmen and Gary argue over what kind of care he should be receiving, and who is going to supply that care. As Valentino draws near death, Mary Carmen finds out she is carrying Valentino's baby. The three take stock of themselves and their relationships with one another.

The film takes Aldous Huxley's novel of the same name and "modernizes" it, setting it in a large metropolitan area much like the cities of today, albeit a bit cleaner, brighter, and free of poverty and crime. The plot centers on Bernard Marx, a high-level "Alpha" executive at the Department of Hatcheries and Conditioning, and on his relationship with Lenina Crowne, a schoolteacher who is responsible for educating the children grown within the building — a dual role, for as well as teaching them out of textbooks, she is also in charge of the sleep-teaching machines that condition the children at night. Marx and Lenina have been seeing each other almost exclusively for a number of months, a practice that is beginning to attract unwanted attention from some in the strictly promiscuous society. On top of this, Marx is increasingly coming under scrutiny by his boss (the Director of Hatcheries and Conditioning) for his wild theories on human psychology and mind control.

Anthony "Tick" Belrose (Hugo Weaving), using the drag pseudonym of Mitzi Del Bra, is a Sydney-based drag queen who accepts an offer to perform his drag act at Lasseter's Hotel Casino Resort managed by a female friend named Marion in Alice Springs, a remote town in central Australia. After persuading his friends and fellow performers, Bernadette Bassinger (Terence Stamp), a recently bereaved transsexual woman, and Adam Whitely (Guy Pearce), a flamboyant and obnoxious younger drag queen who goes under the drag name Felicia Jollygoodfellow, to join him, the three set out in a large tour bus, which Adam christens "Priscilla, Queen of the Desert" for a four-week run in the Australian Outback town.
