The film begins in a little Indian village, where we are introduced to Guru Pitka talking into his recorder which allows him to sound like a man, a woman, or Morgan Freeman. He is the #2 Guru in the world, next to Deepak Chopra. A flashback shows us that Pitka is an orphan of missionaries in India. Both Pitka and Chopra were taught by Guru Tugginmypudha. Pitka says he wants to become a Guru so that girls will love him, so Tugginmypudha puts a chastity belt on him until he can learn that loving himself is more important than being loved by others.

In New York City's Central Park, people begin committing mass suicide. Initially believed to be caused by a bio-terrorist attack using an airborne neurotoxin, the behavior quickly spreads across the northeastern United States. Elliot Moore, a high school science teacher in Philadelphia, hears about the attacks and decides to go to Harrisburg by train with his wife, Alma. They are accompanied by his friend Julian and Julian's eight-year-old daughter Jess. Julian's wife is stuck in Philadelphia but is expected to meet them in Harrisburg. The train loses all radio contact en route and stops at a small town. They receive word that Philadelphia has been attacked by the toxin and Julian's wife was not able to get on the train to Harrisburg. Julian decides to go look for her, leaving his daughter with the Moores while he hitches a ride. However, the town has already been hit by the toxin. Succumbing to it, the driver and Julian both commit suicide.

On his wedding night in 1953, an errant champagne cork renders college engineering professor Robert Kearns (Greg Kinnear) almost completely blind in his left eye. The film begins as married New York City businessman Frank Herlihy (Greg Kinnear) is accidentally killed while trying to buy an apartment for his mistress. Shortly afterward, cynical dentist Bertram Pincus (Ricky Gervais) has a near-death experience while under general anesthetic during a colonoscopy. When he recovers, he is able to see and communicate with ghosts who populate the area. The ghosts annoy Bertram by asking him to help them with personal business that was left unfinished when they died. Frank promises to keep the other ghosts away if Bertram will break up an engagement between Frank's widow Gwen (Leoni), a professional Egyptologist, and Richard (Billy Campbell), a human-rights lawyer who Frank says is dishonest. Bertram eventually agrees to the deal and tries to woo Gwen away from Richard. Bertram's past rudeness to Gwen makes this difficult, but he attracts her interest by analyzing the teeth of a mummified Egyptian Pharaoh that she has been studying.

The film opens with a slight introduction from the main hero of the film, Underdog who introduces the audience to himself, his arch-nemesis Simon Barsinister, and Barsinister's henchman Cad. The film then enters into Capital City Town Hall, where a beagle on the police bomb squad sets off a false alarm after smelling a large ham (in some boxes near where the mayor was giving a speech). After being shunned by his fellow police canines, the beagle leaves in shame and is abducted off the street by Cad, who takes him to Simon's lab. Simon is bitter against the mayor, who has turned down his proposal for a type of genetic experimenting for the city, and plans to prove him wrong by testing his new serum on the beagle. However, Underdog escapes and runs amok in the lab, causing a fire and accidentally becoming exposed to a variety of DNA concoctions and chemicals that Simon developed, which modifies his DNA and gives him the strength to break through the metal door to the lab. After witnessing his work's "success" Simon is stunned when a burning monitor falls onto him, and Cad rushes to his aid.
