The film tells a funny story about three fantastic and fearless female friends. Karen (Joyce Jimenez) is the smart and fashionable one among the three. She works for a girls magazine, and later catches her boyfriend, Artie (Rafael Rosell), in bed with another woman, Greta (Jenny Miller). Love (Assunta De Rossi) is the cousin of Karen. She works in the cosmetics section of a department store, and dreams of becoming a beauty queen someday, thus, making her a regular contestant of beauty competitions. Yolly (Ai-Ai de las Alas) is the businesswoman of the group. She is into the buy and sell business, wherein she buys clothes from ukay-ukay only to sell them as brand new and original. The story runs when Love joins the Diyosa ng Kagandahan 2003 contest, where they find out that her roommate, Greta, is the new girlfriend of Karen's philandering boyfriend, Artie, and the kinakapatid of Yolly's boyfriend policeman, SPO Pablito (Carlos Agassi), who dumped her. They also see this girl cuddling up with one of the judges of the contest, George Collins (Carlo Maceda). Karen, Love and Yolly tire out all their efforts for Love to win the contest and teach their ex-boyfriends a valuable lesson.

Tere Sanchez (Cherry Pie Picache) is an accountant who is in her early forties, single, and not especially happy about it. Tere is an excellent cook, and often throws dinner parties for her friends, and the arrival of her old friend Lorna (Sol Oca) from Manila is all the reason she needs to invite her friends over for a feast. Mike (Christopher De Leon) is a former political journalist who is now enjoying the fruits of a lucrative career as a newspaper editor, but he wonders if he left his principles behind along the way; he's also feeling unfulfilled in his marriage to Gigi (Susan Valdez-LeGoff). Gerry (Ricky Davao) is an advertising copywriter who is afraid to tell his friends and family that he's gay, though circumstances may well drive him out of the closet. Raul (Paolo Montalban) is a good-looking womaniser who prefers to date Caucasians, and lacks a certain amount of emotional maturity. And Marissa (Dina Bonnevie) is on the surface a well-to-do social butterfly; however, deep inside she's woefully insecure, and is afraid to confront her boyfriend Sam (Randy Becker) about his constant infidelity. Meanwhile, Lorna confesses that while she's married to a wealthy man in Manila, she's terribly unhappy, and is considering staying in America as an illegal alien.

Agnes is a meek and dependent girl living with her long-time boyfriend Jerry. When she is invited to live with her estranged mother in California, Jerry pushes her to go through with the trip despite her protests, for him to focus on his work and build his career as an advertising executive. Agnes struggles to survive in the US and tries several times to return to the country, but Jerry insists on prolonging her stay as he struggles to save up for their marriage and support his parents and siblings. He encourages her to get a job and Agnes starts working as a caregiver. Agnes tries to convince Jerry to let her return again and reasons that she has the money to support herself but Jerry insists that as the man of the relationship, he should be capable of supporting her. Jerry starts to avoid Agnes, stressing Agnes out more. The long distance puts a strain on their relationship and ultimately ends when a scared and distraught Agnes is unable to contact Jerry after being put in danger one night. Time passes and Agnes becomes more independent and outgoing, working hard until she lands a job as a real estate agent. Meanwhile, Jerry travels to the US and visits Agnes, with the intention of getting her back. Jerry is overwhelmed by the cultural difference of the US and the Philippines, Agnes' success in her career, and the changes in Agnes herself. Despite this, they still manage to mend their relationship, but Jerry accepts that he is simply unsuited for the kind of lifestyle they have in the US. He leaves Agnes and decides to come home.
