In the kingdom of Malarpuri, the queen (P. Kannamba) delivers a male child. Her brother Naganathan (M. N. Nambiar) replaces it with a dead child with help from a maidservant, and informs king Varaguna Pandiyan (Sadasiva Rao) that the newborn is dead. To Naganathan's surprise, the queen gives birth to another boy soon after delivering the first. Minister Gunaseelar arrives by then and the king is informed of the birth of two children, one dead and one alive. Everyone is happy that at least one is alive. Naganathan decides to kill the first infant and hands it over to his servant Somappa (Stunt Somu). However, Somappa and his wife raise the prince in a forest.

The film tells the story of twin sisters who get separated at birth due to poverty. One is brought up by a wealthy person, and the other by the mother (Lakshmi). A doctor (Gemini Ganesan) comes to treat the ailing mother, and the mother's daughter falls in love with him, of which the doctor is unaware. On her deathbed, the mother reveals to her daughter the existence of the twin. The young woman goes in search of her sister and by sheer chance, both travel in the same train. The train has an accident and the wealthy sister loses her memory and is presumed dead.

"Sikkal" Shanmugasundaram (Sundaram) (Sivaji Ganesan) is a devoted Nadaswaram player, but is short-tempered and sensitive. He meets Mohanambal (Mohana) (Padmini), a Bharatnatyam dancer, and they fall in love with each other. Sundaram's ego prevents him from developing his relationship with Mohanambal, who asks him to play a Thillana on his Nadaswaram while she dances. Mohanambal's mother Vadivambal (C. K. Saraswathi), an influential and mercenary woman, wants her daughter to marry a wealthy man so that she can settle down in life properly. Blinded by love, she disobeys her mother, creating a rift between them. Unaware of her feelings, Sundaram decides to leave the country along with Karuppayi (Manorama), a folk dancer whom he considers to be his sister. Mohana calls him a coward and challenges him to stay, which he readily accepts. In a temple, Sundaram plays the Thillana and Mohana dances. Sundaram, impressed with Mohana's performance, crowns her the title "Thillana Mohanambal". Suddenly Nagalingam (E. R. Sahadevan), a landlord who wanted to marry Mohana, turns up to stab Sundaram with a poisoned knife out of jealousy. He came to the temple to kill Mohana.

Aruna lives a wealthy lifestyle with her paternal grandfather. She has come of marriageable age and he wants her to get married to a young man named Pran. But Aruna finds him possessive, controlling, and hot-tempered, and will not have anything to do with him, so she decides to run away. Her grandfather asks Pran to search everywhere for her, and Pran takes an oath that he will not return home until he finds her. He does find her and brings her back home. Shortly thereafter, her grandpa develops complications and passes away, leaving her in the care of Pran and a servant named Harriya. When Pran offers to marry her, she refuses and tells him that she loves another man, Ashok, she met when she ran away, the one who lives in an abandoned bus in the country. When they locate Ashok, he is unable to recognize her, and is in the company of a beautiful street dancer, Roopa, who he hopes to get married to soon.

Sundar (Sivaji Ganesan) and Uma (Padmini) are classmates who fight often. Sundar, however, is very much in love with Uma, and when they perform in a dance drama competition at Madurai, and later move on to Kodaikanal, he expresses this love. Zamindar of a village has no children and adopts a boy from an Orphanage. The boy grows up as Anandarao (NTR) and returns to the village after higher education. Zamindar, while dying, hands over the estate affairs to Ramaiah (Doraiswamy), who gains good reputation and prestige. He has a son Satyam (ANR) and a daughter Sundaramma (Savitri). Anandrao feels jealous about Ramaiah. Chalapathi (Ramana Reddy) and Pitchaiah (Chadalavada) adds to his jealousy. He insults Ramaiah and stops the donation programme running on the name of Zamindar. Satyam as an unemployee joins as Bus conductor and Ramaiah dies of helplessness. Anandrao purchases the Bus and dismisses Satyam from the job. Chalapathi insults Sundaramma and angered Satyam beats him and goes to jail. Chalapathi and Pitchaiah steal the adoption papers from Anandrao and tries to dismiss him. He joins the Annadana Samajam. This brings about The Change (Parivartana) in him and repents for the injustice done to the Ramaiah family. Sundaramma recognizes the change in his mindset and starts loving him. Satyam recognizes the love affair between them, and recollects the documents from Pitchaiah and provides them in the court. Chalapathi and Pitchaiah are arrested and Anandrao and Sundaramma married happily.
