Set in 1910, the film's main character is Ben Harvey (patterned after Ben Hecht): serious about seeing the world, he leaves his home for Chicago, where he meets a woman named Lil, who in reality is the Madam of the bordello Ben mistakes for a boarding house. In 1593 London, William Shakespeare is a sometime player in the Lord Chamberlain's Men and poor playwright for Philip Henslowe, owner of The Rose Theatre. Shakespeare is working on a new comedy, Romeo and Ethel, the Pirate's Daughter. Suffering from writer's block, he has barely begun the play, but starts auditioning players. Viola de Lesseps, the daughter of a wealthy merchant, who has seen Shakespeare's plays at court, disguises herself as "Thomas Kent" to audition, then runs away. Shakespeare pursues Kent to Viola's house and leaves a note with the nurse, asking Thomas Kent to begin rehearsals at the Rose. He sneaks into the house with the minstrels playing that night at the ball, where her parents are arranging her betrothal to Lord Wessex, an impoverished aristocrat. While dancing with Viola, Shakespeare is struck speechless, and after being forcibly ejected by Wessex, uses Thomas Kent as a go-between to woo her. Wessex also asks Will's name, to which he replies that he is Christopher Marlowe.

In 1997, Sarah Huttinger (Jennifer Aniston), an obituary and wedding announcement writer for The New York Times, travels to Pasadena, California, for her sister Annie's (Mena Suvari) wedding, accompanied by her fiancé Jeff Daly (Mark Ruffalo). At a pre-wedding party, Sarah learns from her grandmother Katharine (Shirley MacLaine) that her mother Jocelyn ran off to Cabo San Lucas to spend time with her prep school classmate Beau Burroughs (Kevin Costner) the week before her wedding to Sarah's father Earl (Richard Jenkins). The film revolves around three characters who work in television news. Jane Craig (Hunter) is a talented, neurotic producer whose life revolves around her work. Chicago Examiner reporter Hildebrand "Hildy" Johnson (Jack Lemmon) has just quit his job in order to marry Peggy Grant (Susan Sarandon) and start a new career, when convict Earl Williams (Austin Pendleton) escapes from death row just prior to his execution. Infuriated at being told to write one final column after being laid off from her newspaper job, Ann Mitchell (Barbara Stanwyck) prints a letter from a fictional unemployed "John Doe" threatening suicide on Christmas Eve in protest of society's ills. When the letter causes a sensation among readers, and the paper's competition suspects a fraud and starts to investigate, editor Henry Connell (James Gleason) is persuaded to rehire Mitchell, who schemes to boost the newspaper's sales by exploiting the fictional John Doe. From a number of derelicts who show up at the paper claiming to have written the original letter, Mitchell and Connell hire John Willoughby (Gary Cooper), a former baseball player and tramp in need of money to repair his injured arm (by Bonesetter Brown), to play the role of John Doe. Mitchell starts to pen a series of articles in Doe's name, elaborating on the original letter's ideas of society's disregard for people in need.
