A zombie outbreak erupts in metropolitan areas around the world; those bitten by the creatures become zombies themselves within twelve seconds. Retired UN investigator Gerry Lane, his wife Karen, and their two daughters manage to escape the outbreak in Philadelphia and take shelter with a family in Newark while waiting for special evacuation the next day, thanks to Gerry's friend UN Deputy Secretary-General Thierry Umuntoni. They find a shelter for the night while waiting for a rescue helicopter. Their hosts choose not to accompany them when they flee the next day, and are subsequently bitten and changed by zombies. Their son, Tommy, manages to escape, and Gerry's family takes him in. They are flown to an offshore U.S. Navy carrier group near New York where Umuntoni is overseeing the remaining worldwide governments' reactions.

Alice Bowman (Meg Ryan) moves to the (fictional) South American country of Tecala because her husband, Peter (David Morse), has been hired to assist with building a dam. Psychotherapist Claire Summers (Anne Hathaway) is called upon by her supervisor (Andre Braugher) to treat a group of five survivors of a recent plane crash; passengers Dean (Ryan Robbins), Norman (Don Thompson), Shannon (Clea DuVall), Eric (Patrick Wilson) and flight attendant Janice (Chelah Horsdal). Claire talks to Eric in the hospital, who seems unusually upbeat. When Claire holds a counselling session for the survivors (except Eric, who decided not to come), she receives different recounts, with some not even remembering certain parts of the crash. Afterwards, Claire meets up with Mr. Arkin (David Morse), who is seemingly an official from the airline to confirm which parts of their recounts are true. He however dismisses her, claiming that the crash was the pilot's fault. She then proceeds to Eric's home for a private counselling, with Eric becoming attracted to her. Claire becomes suspicious of him when he reveals personal information about her, including how he knows what type of coffee she likes and asking whether she has a sister.

In December 1941, just before the USA's entry into World War II, an American agent from the Naval Intelligence Office (Cusack) arrives in Shanghai to find his friend Conner (Jeffrey Dean Morgan) was recently murdered. Determined to find out who killed Conner and why, he begins working at the Herald using the alias Paul Soames, a Nazi-sympathizer cover he used while stationed in Berlin. He meets Anthony Lan-Ting (Chow Yun-Fat) an influential crime lord and Captain Tanaka (Ken Watanabe) at the German embassy in Shanghai, during an invitation only event. He later befriends Anthony when he saves him during an attack on Japanese officers by the Chinese Resistance at a night club.
