David Gordon (John Cusack), a popular science fiction author, was widowed when his wife Mary died as they were trying to adopt a child. The film tells the story of Orton and Halliwell in flashback, framed by sequences of Lahr researching the book upon which the film is based with Orton's literary agent, Peggy Ramsay. Orton and Halliwell's relationship is traced from its beginnings at the Royal Academy of Dramatic Arts. Orton starts out as the uneducated youth to Halliwell's older faux-sophisticate. As the relationship progresses, however, Orton grows increasingly confident in his talent while Halliwell's writing stagnates. They fall into a parody of a traditional married couple, with Orton as the "husband" and Halliwell as the long-suffering and increasingly ignored "wife" (a situation exacerbated by Orton's unwillingness, in 1960s England, to acknowledge having a male lover). Orton is commissioned to write a screenplay for the Beatles and Halliwell gets carried away in preparing for a meeting with the "Fab Four", but in the end Orton is taken away for a meeting on his own. Finally, a despondent Halliwell kills Orton and commits suicide.

Marty Faranan (Farrell) is a struggling writer in Los Angeles, California who dreams of finishing his screenplay, Seven Psychopaths. Marty's best friend, Billy Bickle (Rockwell), is an unemployed actor who makes a living by kidnapping dogs and collecting the owners' cash rewards for their safe return. His partner-in-crime is Hans Kieslowski (Walken), a religious man with a cancer-stricken wife, Myra. Billy helps Marty with Seven Psychopaths, suggesting he use the "Jack of Diamonds" killer, perpetrator of a recent double murder, as one of the seven "psychopaths" in his script. Marty writes a story for another psychopath, the "Quaker", who stalks his daughter's killer for decades, driving the killer to suicide and ultimately cutting his own throat to follow him to hell.

Cervantes and his manservant have been imprisoned by the Spanish Inquisition, and a manuscript by Cervantes is seized by his fellow inmates, who subject him to a mock trial in order to determine whether the manuscript should be returned.
