It was notable for containing the first film role of future star Bruce Cabot, an uncredited bit part (which is an interesting observation since it is a lost film and therefore cannot be examined to identify uncredited players!)

In New York Harbor, Carl Denham, famous for making wildlife films in remote and exotic locations, charters Captain Englehorn's ship Venture for his new project, but he is unable to secure an actress for a female role he reluctantly added. Denham searches the streets of New York for a suitable woman. He meets penniless Ann Darrow and convinces her to join him for the adventure of a lifetime. The Venture quickly gets underway. The surly first mate, Jack Driscoll, gradually falls in love with Ann. After weeks of secrecy, Denham finally tells Englehorn and Driscoll that their destination is Skull Island, an uncharted island shown on a map in Denham's possession. Denham speaks of something monstrous there, a legendary entity known only as "Kong".

Late one night, young David MacLean (Jimmy Hunt) is awakened by a loud thunderstorm. From his bedroom window, he sees a large flying saucer descend and disappear into the sandpit area behind his home. After rushing to tell his parents, his scientist father (Leif Erickson) goes to investigate David's claim. When his father returns much later in the morning, David notices an unusual red puncture along the hairline on the back of his father's neck; his father is now behaving in a cold and hostile manner. David soon begins to realize something is very wrong: one-by-one he notices certain townsfolk are acting in exactly the same way. Through his telescope, David sees child neighbor Kathy Wilson walking in the sandpit, when she suddenly disappears underground. David flees to the police station for help, and he is eventually placed under the protection of health-department physician Dr. Pat Blake (Helena Carter), who slowly begins to believe his crazy story.

The film follows a pilot and crew of a World War II-era Douglas C-47 Skytrain (the military version of the DC-3) who try to survive after a forced emergency landing in the uncharted wildlands near the Quebec-Labrador border. The pilot, Dooley (John Wayne), is a former airline pilot, who, like many others, was pressed into duty hauling war supplies across the northern route to England. The film opens with a brief scene serving as its introduction. A man is being caressed by feminine hands. The next shot includes the face of the woman, Tarantella (Tandra Quinn). A brief kiss between her and the man, ends with his lifeless body falling down. A disembodied voice asks the audience "Have you ever been kissed by a girl like this?" The narrative properly begins in a desert. A narrator (Lyle Talbot) mocks the overblown ego of humanity, a race of puny bipeds which claims to own planet Earth and every living thing on it. Yet, they are outnumbered by the insects, and the Hexapods are likely to survive longer that the humans. The narrator then claims that when men or women venture off "the well beaten path of civilization" and deal with the unknown, the price of their survival is the loss of their sanity.
