Murli Prasad Sharma alias Munna Bhai (Sanjay Dutt), a gangster who is in love with the voice of Janhavi (Vidya Balan), a radio jockey, devises a plan to meet her when she announces a quiz contest featuring the life and beliefs of Mahatma Gandhi, to be aired on 2 October, the birthday of Gandhi. To prepare for the contest, Munna's sidekick Circuit (Arshad Warsi) kidnaps and bribes a group of professors to provide the answers for Munna. After winning the contest, Munna is granted an interview with Janhavi wherein he presents himself as a professor of history and a Gandhi specialist. Janhavi subsequently asks Munna to present a lecture on Gandhi to a community of senior citizens who live in her home, called the 'Second Innings House.' To prepare for this event, Munna engages in intense study at a Gandhi institute. For three days and nights (and without food or sleep), Munna reads about the life and ideologies of Gandhi.

Murli Prasad Sharma (Sanjay Dutt), nicknamed "Munna Bhai" (literally "Brother Munna") is a bhai or gunda: a crime don in the Mumbai underworld. Given that his father had wished him to be a medical doctor, he creates the faux Sri Hari Prasad Sharma Charitable Hospital (named after his father) and pretends to live in accordance with this wish whenever his father (Sunil Dutt) and mother (Rohini Hattangadi) visit him in Mumbai.

A humanoid alien (Aamir Khan) lands on Earth naked on a research mission in Rajasthan, but is stranded when the remote control of his spaceship is stolen, though he manages to get the thief's cassette recorder, a National Panasonic RQ-565D. On the same day in Bruges, Jaggu (Anushka Sharma) meets a man named Sarfaraz (Sushant Singh Rajput) and falls in love with him. Jaggu's father (Parikshit Sahni) objects to their relationship because Sarfaraz is a Pakistani Muslim, while Jaggu is a Hindu. He consults godman Tapasvi Maharaj (Saurabh Shukla) who predicts that Sarfaraz will betray Jaggu. Determined to prove them wrong, Jaggu proposes to Sarfaraz. Farhan Qureshi (R.Madhavan) and Raju Rastogi (Sharman Joshi) are students at the Imperial College of Engineering (ICE), a highly regarded Indian engineering college. Farhan's passion is wildlife photography, but he pursues an engineering degree to appease his father. Raju chooses engineering with hopes of improving his family's financial situation, but his lack of self-confidence results in poor grades. Their third roommate and friend, Ranchodas 'Rancho' Shamaldas Chanchad (Aamir Khan), has immense interest in robotics, and invents in his spare time. Rancho regularly faces the wrath of the college's director, Dr. Viru Sahastrabuddhe (Boman Irani), known as "Virus", when he gives creative and unorthodox answers in class. He lives by the philosophy that one should pursue excellence, as when it is achieved, success follows. Virus is shown as an extremely strict person—so strict that he caused his own son's suicide by putting him under excess pressure. The star student of their class is Chatur Ramalingam (Omi Vaidya) nicknamed "Silencer", known for his incredible techniques of memorization but more notorious for his silent but malodorous flatulence in addition to his arrogant attitude. Catering for them and the other college students' needs is Man Mohan, nicknamed "Millimeter".

The film begins with a drunk Rahul Dewan (Rahul Siddhartha), racing in an Sports Utility Vehicle against a car, after he is seduced by a woman in that car. He loses control and crashes into a wall on the footpath by the road. The film cuts to a court scene where Jagdish Tyagi aka Jolly (Arshad Warsi), a Lawyer defending a surgeon against a Public interest litigation (PIL) filed against him after the surgeon had his son operate upon a patient in his pursuit of Guinness Book of World Records. Jolly loses the case but decides to move to the capital Delhi to try his luck. He happens to see prominent criminal lawyer Tejinder Rajpal (Boman Irani) defending Rahul Dewan in a sessions court. Rahul Dewan is accused of causing death under the influence of alcohol. Rajpal convinces the judge that prosecution had no evidence to implicate Rahul Dewan and he was being deliberately targeted by the media as he was from a rich family, resulting in Dewan's acquittal.
