After having killed the notorious extortionist Mayil Vaaganam in the first film, Duraisingam (Suriya) has gone undercover after meeting the Home Minister and is working as an NCC officer in a school in Thoothukudi. The only people who know about this operation are the Chief Minister and Home Minister apart from himself. Sathya (Hansika Motwani) is a student who falls for Duraisingam though he is waiting to marry Kavya (Anushka Shetty), his love interest from the previous film. Susai (Santhanam), a local who works in the same school shows Duraisingam parts of Thoothukudi and also tells him about the rogue elements dominating the area helping him in his surveillance. He requests the Home Minister to transfer Erimalai (Vivek) and another cop for assisting him. Sathya's moves are frequently repulsed by Duraisingam who learns that Sathya is the niece of Thangaraj (Rahman). Bhai (Mukesh Rishi) and Thangaraj, are two criminals who control the area. Bhai and Thangaraj have connections with an international gangster and drug lord Danny (Danny Sapani).

Masi (Nakul) is an orphan living in a lower middle class colony. The story revolves around three roommates, journalist J K Boss(Arya), photographer Nagaraj(Santhanam) and Cheenu (Premji) leading an unkempt and debt-ridden life in a shoddy apartment. J K's ditsy fiancée, Madhumita, is an air hostess who agrees to deliver a package for Vladimir Dragunsky (Kim Bodnia) to Nassar, without realizing its contents or that Nassar is a gangster. Madhumita asks J K to deliver the package. JK, in turn, asks Nagaraj to do so. But Nagaraj is unable to do so as he is experiencing 'Chennai Belly'. Nagaraj hands Madhumita's package to Cheenu for delivery to Nassar, along with a package containing his stool sample for delivery to Nagaraj's doctor. Cheenu mixes up the two packages which makes Nassar furious and he starts the investigation by interrogating Vladimir.

The film starts with a flashback. ACP Varadhanayaka (Sudeep) is a sincere police officer happily married and lives with his parents and younger brother Hari (Chiranjeevi Sarja). Hari is a college student and he falls in love with his classmate Sirisha (Nikeesha Patel). Enter Section Shankar (P. Ravi Shankar) the villain who is notorious for settlements and land deals. Surya is accused of shooting Mythilli. Meghala is on the case of Mythilli's death.

The movie starts with Sugan(Karthi) and his friend Parasuram(Premgi Amaren ) being chased by cops.
