Bernie Reubens (Gregg Sulkin), a young Jewish boy, is about to have his bar mitzvah. The picaresque story follows its protagonist, Pasqualino (Giannini) who, as a dandy and small-time hood in Naples, to save the family honour, is sent to prison for killing a pimp (and then dissecting the victim and placing the body in suitcases) who had turned Pasqualino's sister into a prostitute. Convicted and sent to prison, Pasqualino succeeds in being transferred to a psychiatric ward. Desperate to get out, he volunteers for the Italian Army, but then somewhere in Germany he deserts with a comrade. They are captured and sent to a concentration camp. There, in a bid to save his own life, Pasqualino decides to survive by providing sexual favors to the obese and ugly female commandant (Stoler). His plan succeeds, except for the fact that he is then put in charge of the barracks as a kapo, and is obliged to select six men to be killed under the threat that if he doesn't do so, they will all be killed. Pasqualino ends up executing the soldier with whom he was captured and being responsible for the death of another fellow prisoner, a Spanish anarchist. At the war's end, upon his return to Naples, Pasqualino discovers that his seven sisters, his fiancée and even his mother have all survived through prostitution.

The movie starts off with a man, named Schlomo (Lionel Abelanski), running crazily through a forest, with his voice playing in the background, saying that he has seen the horror of the Nazis in a nearby town, and he must tell the others. Once he gets into town, he informs the rabbi, and together they run through the town and once they have got enough people together, they hold a town meeting. At first, many of the men do not believe the horrors they are being told, and many criticize Schlomo, for he is the town lunatic, and who could possibly believe him? But the rabbi believes him, and then they try to tackle the problem of the coming terrors. Amidst the pondering and the arguing, Schlomo suggests that they build a train, so they can escape by deporting themselves. Some of their members pretend to be Nazis in order to ostensibly transport them to a concentration camp, when in reality, they are going to Palestine via Russia. Thus the Train of Life is born.

The film opens with on-screen text stating: "A true story". It is August 1941, and Nazi Einsatzgruppen are sweeping through Eastern Europe, systematically killing Jews. In 1941, SS colonel Hans Landa, the "Jew Hunter", interrogates French dairy farmer Perrier LaPadite as to the whereabouts of the last unaccounted-for Jewish family in the neighborhood. Under threat of continued harassment from the Germans, LaPadite reveals that the Dreyfus family is hidden under the floor. Landa orders SS soldiers to shoot through the floorboards and kill the family. All are killed except Shosanna, who escapes and flees through the field. Landa watches her go, deciding not to shoot her at the last second.

Jonathan Foer, (Elijah Wood) a young American Jew, goes on a quest to find the woman, Augustina, sister of Lista (Laryssa Lauret), who saved his grandfather during the Holocaust in a small Ukrainian town called Trachimbrod that was wiped off the map when the Nazis liquidated Eastern European shtetls. His guides are a cranky, anti-semitic grandfather (Boris Leskin); his deranged Border collie named Sammy Davis Jr. Jr.; and his over-enthusiastic grandson, Alex (Eugene Hutz), whose fractured command of English, passion for American pop culture, and constant chatter threaten to make the worst of every situation. The guides are not very knowledgeable about the subject of finding Jews, and usually just attempt to scam them by taking them on long journeys, however after hearing about Jonathan's compelling story, they decide they actually want to help him. After traveling through much of rural Ukraine they eventually find Augustina's sister, who leads them to where Augustina was killed by Nazi soldiers after her father refused to spit on the Torah. The grandfather kills himself after it was revealed he was Jewish and managed to survive the war himself. His suicide was portrayed as more of a relief than a tragedy. Jonathan returns home after saying farewell to his new friend Alex.
