The film was written by Albert DeMond (titles), John Grey (story), J.A. Howe (story), Lex Neal (story), and Howard Emmett Rogers (story) with uncredited assistance from Al Boasberg and Paul Girard Smith. The film was directed by Ted Wilde, the last silent film to be directed by him, and was shot in both Hollywood, and on location in New York City.
The plot revolves around Harold 'Speedy' Swift's attempts to save the last horse-drawn streetcar in New York. The film contrasts the speed of life of the contemporary city with the pace of yesteryear, represented by this non-motorized mode of transport. Yankees star Babe Ruth plays one of 'Speedy's' hapless passengers.
There is a scene in the film where Speedy is seen giving the finger to himself while looking in a distorted mirror which may be the earliest motion picture depiction of that gesture.

Novelist Larry Donner (Billy Crystal) struggles with writer's block due to his resentment towards his ex-wife Margaret (Kate Mulgrew), who stole his book and garnered mainstream success and critical acclaim with it. Owen Lift (Danny DeVito) is a timid, middle-aged fellow who still lives with his overbearing, abusive and paranoid mother (Anne Ramsey). Owen fantasizes about killing his mother but can't summon the courage to bring his desires into fruition. As a student in Larry's community college writing class, Owen is given advice by Larry to view an Alfred Hitchcock film to gain some insight into plot development for his mystery stories. He sees Strangers on a Train, in which two strangers conspire to commit a murder for each other, figuring their lack of connection to the victim will, in theory, establish a perfect alibi. Having overheard Larry's public rant that he wished his ex-wife dead, Owen forms a plan to kill Margaret, believing that Larry will, in return, kill his mother.

Coming to the assistance of a nanny who is almost killed during a bungled hit-and-run assassination attempt, Richard Hannay (More) is surprised to find that there is no baby in her pram. Curious, he meets her at the Palace Music Hall where she has gone to see the act of Mr Memory (James Hayter). Afterwards, she goes back to Hannay's flat with him, where she reveals that she is a spy working for British Intelligence following a group called "The Thirty-Nine Steps"; all they know about their elusive leader is that he is missing the tip of a finger. The Thirty-Nine Steps are in possession of a set of top-secret plans for "Boomerang", a British ballistic missile project that could tip the balance of power in Europe. She tells Hannay that she must leave for Scotland immediately, but while Hannay is out of the room, she is killed by two hitmen.

On Christmas, we meet foster brothers John and Charlie. They work as transit cops patrolling the New York City subway. When a mugging occurs, John and Charlie chase the mugger into a subway tunnel, and all trains traveling in their direction are halted. But their harsh transit captain Donald Patterson, allows the money train to continue. John and Charlie avoid getting hit by the train; however, when transit police at the next station witness the mugger running toward the money train, they shoot him dead. The mugger is revealed to be a young teenage boy, which triggers a brawl between John, Charlie, and the other transit officers. Patterson blames the two for causing his money train to arrive late.

Donald Sinclair (John Cleese), the eccentric owner of The Venetian Resort Hotel Casino in Las Vegas, devises a new game to entertain the high rollers who visit his hotel. Six special tokens are placed in the casino's slot machines, and the winners are gathered together and told that $2 million in cash is hidden in a duffel bag in a train station locker in Silver City, New Mexico, 563 miles southeast of Las Vegas. Rahul Mithaiwala (Shahrukh Khan) is a forty-year-old bachelor living in Mumbai. His parents died in a car accident when he was eight years old, so his grandparents raised him. His grandfather (Lekh Tandon) owns a chain of confectionery shops where Rahul has been a dedicated worker helping his grandfather. Two of Rahul's friends convince to finally take a vacation and travel with them to Goa. Rahul's grandfather dies on the day of the 100th birthday celebration. His grandmother tells Rahul that his grandfather wanted his ashes to be divided into two parts—one of each to be immersed in the Ganges and Rameswaram. She asks Rahul to take the ashes to Rameswaram and disperse them. While he accepts her request, Rahul and his friends plan to dump the ashes at Goa but his grandmother wants to see him off, forcing Rahul to travel by train. He books a single ticket on the Chennai Express, planning to quickly exit and meet his friends along the way. However, as he tries to leave the train, he notices a young woman running to board it. He helps her and then four large men board the moving train, missing his chance to debark.
