Shelley Darlington (Anna Faris) is an aspiring Playboy Playmate living the life of luxury in the Playboy Mansion. The day after her twenty-seventh birthday, she awakes to find a note, seemingly from Hugh Hefner, asking her to pack up and leave. Bartleby Gaines (Justin Long) is a persuasive senior from William McKinley High School in Wickliffe, Ohio, who, among other pranks, creates fake IDs. His gifts do not extend to grades, however, and he receives rejection letters from all of the colleges to which he applies, including those with high acceptance rates. In an attempt to gain approval from his strict father (Mark Derwin), Bartleby creates a fake college, the South Harmon Institute of Technology (S.H.I.T.). His best friend, Sherman Schrader III (Jonah Hill), who has been accepted into his father's prestigious alma mater Harmon College, aids Bartleby and fellow rejects Rory Thayer (Maria Thayer), who only applied to Yale University; Hands Holloway (Columbus Short), who lost his athletic scholarship after an injury; and Glen (Adam Herschman), who failed his SAT exam. To make the "college" seem legitimate, Bartleby convinces Sherman to create a functional website for the school.

The mouse and his child are two parts of a single small wind-up toy, which must be wound up by means of a key in the father's back. After having been unpacked, they discover themselves in a toy shop where they befriend a toy elephant and toy seal. The child mouse proposes staying at the shop to form a family, which the other toys ridicule. After breaking due to a fall from the counter, they are thrown in the trash. Outside, they become enslaved by Manny the rat, who runs a casino in the city dump and uses broken wind-up toys as his slave labor force. With the aid of a psychic frog, the mice escape and meet various animal characters on a quest of becoming free and independent self-winding toys. They rediscover the elephant and seal, who are somewhat broken down. Together they manage to form a family and destroy the rat empire.

Megamind is a super intelligent alien who was evacuated from his homeworld as a baby, as was Metro Man; the two fall into paths of super villainy and superheroism respectively and grow up as rivals fighting for control of Metro City. Megamind is consistently defeated by Metroman and is in prison. After using a holographic watch to escape with the aid of Minion, a talking fish with the robotic body of a gorilla, he kidnaps Metro Man's supposed love interest, reporter Roxanne Ritchi and holds her hostage to lure him into a trap. Finding that copper is Metro Man's one weakness, Megamind's plan to obliterate him with a death ray powered by the sun succeeds, and Megamind finally takes over the city. His joy is short lived though, as without a hero to fight, he finds his life has become meaningless. He goes to the Metroman Museum, which was dedicated to him on the day of his death and nearly runs into Roxanne. He uses his holographic watch to disguise himself as the museum's curator Bernard, and she innocently gives him the idea of creating a new superhero to take Metro Man's place. After creating a formula from Metroman's DNA, Roxanne intervenes in his plans and he accidentally injects the serum into Hal Stewart, Roxanne's dimwitted cameraman, who has an unrequited crush on her. Under the guise of his "Space Dad", Megamind tries to mold Hal into a superhero named Titan (misheard by Hal as "Tighten"), as it was the only name he could trademark. Unfortunately, Hal's ambitions are crushed when he sees Roxanne and Megamind as Bernard on a date. However, Megamind's disguise falters during dinner and she rejects him, causing him to lose track of his invisible car which contains the gun capable of removing Hal's powers.

Dave Walsh (Matt Dillon) is a bank employee whose day begins badly when he gets fired from his dream job and dumped by his fiancée Sara Goodwin (Christina Applegate) the day after their engagement party was crashed by his best friend, Jack (Steve Zahn). Jack tries to console him by telling him that it's for the best, that Dave was headed down the wrong path. The next day, Dave goes back to the bank with a gun tucked into in his waistband. He surreptitiously makes his way into his former boss's office, where he pulls out the gun and threatens to end his boss's life. Though intimidated, the boss stands up to Dave, and Dave, deciding he does not want blood on his hands after all, pistol-whips him instead.
