The film explores the tension between two Spokane men who grew up together on the Spokane Reservation in eastern Washington state: Seymour Polatkin (Evan Adams) and Aristotle (Gene Tagaban). Seymour's internal conflict between his Indian heritage and his life as an urban gay man with a white boyfriend plays out in multiple cultures and relationships over his college and early adult years. His literary success as a famed American Indian poet, resulting in accolades from non-Indians, contrasts with a lack of approval from those he grew up with back on the reservation. The protagonist struggles with discomfort and alienation in both worlds.
Seymour returns to the reservation for the funeral of his friend Mouse (Swil Kanim), a violinist, and Seymour's internal conflict becomes external as his childhood friends and relatives on the reservation question his motivation for writing Indian-themed poems and selling them to the mainstream public. The film examines several issues that contemporary American Indians face, including cultural assimilation (both on the reservation and in urban areas), difficult stereotypes, and substance abuse.
In the DVD commentary, Alexie refers to Michelle St. John's character, Agnes Roth, a mixed-race (Spokane/Jewish) woman who moves to the reservation to teach in the school, as "the moral center of the film". Agnes is also an ex-lover of Polatkin's, with the two of them still maintaining a deep friendship.
The film's incidental music was composed by Mohican composer Brent Michael Davids. The violin solos were composed and performed by Swil Kanim, and a number of the actors sing. The film also features Alexie's poetry, and the author's mother served as a language consultant.
The film was made in an experimental and largely non-hierarchical manner, with a predominantly female crew; many scenes were improvised, with biographical details from the lives of the actors as well as the writer/director. This is discussed in detail on the DVD commentary and the behind the scenes documentary included in the DVD release, where Alexie comments that he wanted to make a film that not only discussed his politics, but put them into practice in the making of the film.

A group of dancers congregate on the stage of a Broadway theatre to audition for a new musical production directed by Zach (Michael Douglas). After the initial eliminations, sixteen hopefuls remain. Arriving late is former lead dancer Cassie (Alyson Reed) who once had a tempestuous romantic relationship with Zach but left him to take a job in Hollywood. Ali Rose (Aguilera) moves to Los Angeles after she quits her bar job when her boss refuses to pay her. Once in L.A., she tries and fails at every audition she does until one night, she finds herself unknowingly in a burlesque club when she hears the music on the street. She finds Tess (Cher) and the dancers performing “Welcome to Burlesque” and decides to pursue a career on stage once she meets Jack (Gigandet). Jack refers her to Tess for an audition, but is rejected instantly and ushered out by Sean (Tucci). Instead of leaving, Ali begins serving customers at the club as a waitress, while Tess and Sean observe with Jack asking Tess to give Ali a chance.

Nicky Marotta (Robin Johnson) and Pamela Pearl (Trini Alvarado) are two teenage girls who meet in the New York Neurological Hospital, where they are both being examined for mental illness. Pamela feels lonely and misunderstood, and is neglected and exploited by her father, a prominent and wealthy environmental commissioner running a campaign to "clean up" Times Square. Nicky is a tough-talking street kid with musical aspirations, sent to the hospital for an evaluation after an altercation with the police. Sharing a room, the frank Nicky and the shy Pamela become friends; Nicky tells Pamela that she stages seizures to keep out of juvenile detention. Nicky admires Pamela's poetic spirit; Pamela admires Nicky's forthright attitude, and resents the condescending way in which the doctors treat her. Nicky tells Pamela not to take her meds as they "take your fight away." She also reads a poem that Pamela wrote about her: "Your ribs are my ladder, Nicky/I'm so amazed, I'm so amazed." Nicky is released from the hospital, and later returns, ostensibly for an appointment for her social worker, but really to break Pamela out. Both girls escape from the hospital, steal an ambulance, and hide out in an abandoned Chelsea Piers No.56 on the Hudson River. Pamela tells Nicky, "Everything you do, or you say, is poetry. At least, I think so." The girls seal their devotion to each other with a blood oath and make a pact to scream out each other's names in times of trouble.

A straight bass player in an "all gay band" called The Jerk-Offs, Nick O'Leary (Michael Cera) is a teenager from Hoboken, New Jersey. Nick is still heartbroken after his girlfriend, Tris (Alexis Dziena), broke up with him three weeks, two days, and 23 hours ago, and continues to make for her "breakup" mix CDs. Thom (Aaron Yoo) and Dev (Rafi Gavron), his gay bandmates who both hate Tris, convince Nick to perform at a club because his favorite band— the legendary, publicity-shy indie band Where's Fluffy? —is performing somewhere in New York City that night. Norah Silverberg (Kat Dennings) attends the same high school as Tris, Convent of the Sacred Heart, and though Norah and Tris dislike each other, they have a mutual friend named Caroline (Ari Graynor). Norah has not met Nick, but shares his taste in music based on recovering his mix CDs after Tris discards them in the trash. That night, the three girls end up at Arlene's Grocery on Manhattan's Lower East Side, where The Jerk-Offs are playing. Norah asks Nick to pretend to be her boyfriend to show up Tris. Norah kisses him, unaware that he is the ex-boyfriend of Tris. Caroline gets drunk and Norah wants to take her home, but Nick's bandmates (who see Norah as a potential new girlfriend for him) offer to take Caroline home so Norah and Nick can spend time together trying to find the show.

The film centers on the experiences of several teenagers at Camp Ovation, a summer theatre camp. The campers are a mishmash of youngsters from different backgrounds and dealing with different issues, but all feel most comfortable while performing. School is out, and the campers reunite as they prepare to board the buses to Camp Ovation. Shy, self-conscious Ellen Lucas happily greets her friends, recovering from an embarrassing situation where she desperately tried to land a prom date, and ended up asking her brother. Returning camper Jenna Malloran laments that her parents forced her to have her jaw wired shut in order to lose weight. Upon arrival to Camp Ovation, the kids settle in. Ellen converses with her friend Michael Flores, a gay teenager who was violently beaten by his classmates after showing up to his junior prom in drag. The campers are struck dumb with the arrival of Vlad Baumann, a handsome new camper who is, as a staff member marvels, "an honest-to-god straight boy.
