Jesse and Chester awaken with hangovers and no memory of the previous night. The television is on, showing an Animal Planet program about how animals use twigs and rocks as tools to get food. Their refrigerator is filled with containers of chocolate pudding, and the answering machine contains an angry message from their twin girlfriends Wilma and Wanda as to their whereabouts. They emerge from their home to find Jesse's car missing, and with it their girlfriends' one-year anniversary presents. This prompts Jesse to ask the film's titular question: "Dude, where's my car?"

Investment banker Harold Lee (John Cho) is persuaded by his colleagues Billy and JD to do their work while they leave for the weekend. Elsewhere, Kumar Patel (Kal Penn) attends a medical school interview at the behest of his father but intentionally botches it to prevent getting accepted. Upon arriving home, Harold meets his Latina neighbor Maria (Paula Garcés) but is unable to admit his feelings for her. After smoking marijuana with Kumar, and seeing an advertisement for White Castle, the pair decide to get hamburgers to satisfy their hunger. Traveling to the location of the nearest White Castle, the pair find it has been replaced by "Burger Shack" but are informed that there is another White Castle in Cherry Hill.

The three meet each other for the first time at Mumbai's airport and decide to find a place together. Harold Crick, an agent for the Internal Revenue Service, lives his life by his wristwatch. He is assigned to audit an intentionally tax-delinquent baker, Ana Pascal, to whom he is attracted. On the same day, he begins hearing the voice of a woman omnisciently narrating his life but is unable to communicate with it. Harold's watch stops working and he resets it using the time given by a bystander; the voice narrates, "little did he know that this simple, seemingly innocuous act would result in his imminent death". Worried by this prediction, Harold consults a psychiatrist who attributes the voice to schizophrenia, though they consider that if there really is a narrator, he should visit an expert in literature. Crick visits Jules Hilbert, a literature professor, and relates his story. When Jules recognizes aspects of a literary work in Harold's story, he encourages Harold to identify the author, first by determining if the work is a comedy or tragedy.

Jon (Domhnall Gleeson), an aspiring songwriter, witnesses a man trying to drown himself while walking along the beach of his town. The man is taken to the hospital and Jon talks to Don (Scoot McNairy), who explains the man was a keyboardist in an experimental band, the Soronprfbs, managed by him. Jon mentions that he plays keyboards and is invited to play with them that night. Jon goes along and meets the rest of the band, all of whom are reluctant about Jon, except Frank (Michael Fassbender), the band leader who wears a papier-mâché mask.
