Left off after the events of the first movie The Blob, an oil pipeline layer named Chester (Godfrey Cambridge) returns to his suburban Los Angeles home from the North Pole, bringing with him a small sample of a mysterious frozen substance uncovered by a bulldozer on a job site. Prior to taking the blob to a lab to be analyzed, he places the storage container with the substance in his freezer, but he and his wife (Marlene Clark) accidentally let it thaw, releasing "the Blob". It starts by eating a fly, then a kitten, Chester's wife, and then Chester himself (while he is watching a television broadcast of the film The Blob).

Detective Lucas McCarthy (Lance Henriksen) finally catches the serial killer named "Meat Cleaver Max" (Brion James) who killed over 100 people and watches his execution. McCarthy and the others watching the execution are shocked to see the electric chair send enough voltage through him to physically burn Max's body before finally dying. Max, however, has made a deal with the devil in order to return from the grave and frame Lucas for a series of grisly murders. He also scares the McCarthy family (who have moved into a new house) and the parapsychologist they hire. Lucas' only hope of stopping Max for good is to destroy his spirit before Max destroys his life and family.

Mary Carroll (Lizabeth Scott) inherits her family's ancestral home, located on a small island off Cuba, and, despite warnings and death threats, decides to sail to Havana and take possession of the reputedly haunted castle. Chuck Murray (Bud Abbott) and Ferdie Jones (Lou Costello), gas station attendants, aspire to better jobs waiting tables at Chez Glamour, a high-class nightclub, where Ted Lewis and The Andrews Sisters perform. However, Chuck and Ferdie cause a ruckus and wind up back at the gas station. Gangster "Moose" Mattson (William B. Davidson) brings his car in for servicing, and Chuck and Ferdie are caught inside the vehicle when the gangster speeds off to escape the police. During the chase shots are exchanged, and the gangster is killed by gunfire. However, through a strange clause in his will--which says that whoever is with him when he dies will get whatever he owns--Chuck and Ferdie inherit his rural tavern, the Forrester's Club. Mattson had also given a cryptic clue about a hidden stash of money, stating that he always kept it "in his head," but its existence and location remain a mystery.

After Slip discovers a spiritualist in the neighborhood, he enlists the boys to investigate. They discover that she is a fake and working for Margo the Medium, a radio star who has convinced Sach and Whitey that ghosts exist. They go to her place using Louie as a decoy. While she is busy trying to connect to Louie's dead uncle, the rest of the boys investigate the house. A real ghost, Edgar, befriends Sach and helps him investigate as well. Although the others cannot see Edgar, he does help them in times of crisis and helps them uncover Margo's scam.
