Singing cowboy Gene Autry (Gene Autry) is the foreman of the Circle R Ranch, which has been in the Adams family for generations. The ranchers in the area have enjoyed free grazing rights on the Circle R for years. Recently, the madcap heiress of the ranch, beautiful Jo Adams (Virginia Dale), negotiated the sale of the ranch in order to pay off some of her debts. She accepted a $25,000 down payment, with an option to purchase in 60 days, from unscrupulous cattle broker John Ramsey (George Meeker) who is conspiring with Adams' business manager James Morgan (Harry Stubbs) to buy the ranch and cut off grazing rights to the ranchers.

In the Christmas spirit, Boston Blackie decides to entertain the inmates at his old "alma mater" by bringing a variety show headed by clown Roggi McKay (George McKay). However, Blackie kindheartedly lets her come along. Inspector Farraday (Richard Lane and Detective Joe Mathews (Walter Sande) unexpectedly join the group on the bus, just to keep an eye on Blackie. When Joe manages to escape from prison, by tying Roggi up and putting on his costume and makeup, Farraday suspects Blackie helped him.

Faced with mortgage debts, Professor Nathaniel Billings (Boris Karloff) sells his 18th-century tavern to Winnie Layden (Jeff Donnell), who plans to turn it into a hotel. Billings stipulates as a condition of sale that he is able to continue working in a laboratory in the basement. His housekeeper Amelia Jones (Maude Eburne) and hired hand Ebenezer (George McKay) also continue to work in the inn. Layden is initially unaware of the nature of Billings's experiments in the basement laboratory: he is attempting to use electricity to create a race of superhumans to help the war effort. Layden's ex-husband Bill (Larry Parks) is against the sale, but is too late to stop it, and decides to stay on at the inn for a few days.

When "Diamond" Ed Barnaby (an uncredited Walter Baldwin) is paroled, he sets out to give $100,000 worth of diamonds to his daughter, Betty (Ann Savage). Aware that several shady characters know he has the jewels, he stashes them in a safe deposit box in the Arcade Building. Betty later receives a call asking her to meet him there, but he never shows up. She contacts her father's only real friend: Horatio Black, better known as Boston Blackie. He agrees to help and drops Betty off at the apartment of his wealthy friend, Arthur Manleder (Lloyd Corrigan), for safekeeping. His sidekick, "the Runt" (George E. Stone), has to postpone his wedding to statuesque bubble dancer Dixy Rose Blossom.
