Edith von Felsen est une orpheline d’un bon milieu. Invitée un jour à Londres par sa tante, elle rencontre au cours de son voyage une dame qui manifestement commence à s’intéresser beaucoup à elle. Florence Grange (Clara Pontoppidan), a pretty young Danish woman, is vacationing with her father, her brother, George, and her fiance, Leslie, at a luxurious seaside hotel in Denmark. One evening, while they are sitting in the restaurant, George introduces them to a young American named Andrew Larson (Valdemar Psilander). Andrew, who is a Mormon priest, is quickly attracted to Florence and gives her an "admission card" to a Mormon meeting. Although Florence cares greatly for Leslie (Carlo Wieth), he often neglects her while pursuing other interests. Florence spends much time with the strangely fascinating American, enabling Andrew to indoctrinate her and convince her to go to Utah with him. Partly in passion and partly in hypnotic trance, Florence runs away and meets Andrew at the railway station.

In 1462, Vlad Dracula, a member of the Order of the Dragon, returns from a victory against the Turks to find his wife, Elisabeta, has committed suicide after receiving a false report of his death. Enraged that his wife is now damned for committing suicide, Dracula desecrates his chapel and renounces God, declaring that he will rise from the grave to avenge Elisabeta with all the powers of darkness. In a fit of rage, he stabs the chapel's stone cross with his sword and drinks the blood which pours out of it.

In the Middle Ages, Vlad Tepes (Luke Evans) is the Prince of Wallachia and Transylvania. As a child, he was a princely hostage of the Ottoman Empire and was trained to be a soldier in the Sultan's elite Janissary corps, where he became their most feared warrior. He was called Vlad the Impaler after slaughtering thousands by impaling them on spears. Eventually sickened by his acts, he put aside his past and returned to rule his domains in peace. One day in the forest, Vlad and his soldiers discover a helmet in a stream. Fearing that an Ottoman scouting party is preparing the way for invasion, they follow the stream to a high mountain cave called Broken Tooth Mountain. Upon entering the cave, they discover the ground of the cave is carpeted in crushed bone; and they are attacked in the dark by an unknown creature (Charles Dance) with shining red eyes, as well as inhuman speed and strength. While his men are killed, Vlad cuts the creature of the cave with his sword before being thrown to the ground. The blood on the sword he used to cut the creature dissolves when exposed to sunlight, and the creature does not pursue Vlad into the sunlight at the mouth of the cave. Returning to his castle, Vlad learns from a local monk that the creature is a vampire, who was once a man who summoned a demon from the depths of hell and made a pact with it for dark powers before being tricked by the demon and cursed to remain in the cave forever until he is released by someone who drinks his blood, then drinks the blood of a human. The person is welcome to share his power in exchange for the freedom to escape the cave.

Kathleen Conklin (Taylor), a young philosophy student at New York University, is attacked by a woman (Annabella Sciorra), who tells her "order me to go away" and, when the frightened Kathleen is unable to do so, bites her neck and drinks her blood. Kathleen develops several of the traditional symptoms of vampirism, including aversion to daylight, but the film's main focus is on her moral degradation. It is hinted that vampires become immortal in this film, but the price is an addiction to blood. Vampires are shown repeatedly resorting to the strategy of blaming their victims for not being strong enough to resist them. As one of Kathleen's victims weeps incredulously over the damage, Kathleen coldly informs her: "My indifference is not the concern here - it's your astonishment that needs studying." Eventually Kathleen meets Peina (Walken), who claims to have almost conquered his addiction, and as a result is almost human. For a time he keeps her in his home trying to help her overcome hers, recommending that she read William S. Burroughs' Naked Lunch. At her graduation party, she says "I'd like to share a little bit of what I've learned" she and her victims (now vampires themselves) attack the party goers, participating in a bloody, chaotic vampire orgy.

In 2010 an old man picks up a discarded note dropped by teenage vampire Eleanor Webb, who has taken to writing her life story but then throwing the individual pages to the wind. Realising what she is, the old man invites Eleanor to his house, and tells her that he is ready for death. Eleanor proceeds to kill him and consume his blood. Elsewhere, Eleanor's mother, Clara, is chased from the lap-dancing club where she has been working by Werner, a member of the vampiric Brethren, who demands to know where Eleanor is. Clara decapitates Werner, burns his body and leaves town with her daughter.

In 2009, a genetically re-engineered measles virus, originally created as a cure for cancer, turns into a lethal strain which kills 90% of those it infects, and mutates the remaining 10% into predatory, nocturnal mutants called "Darkseekers" who are extremely vulnerable to sunlight and other sources of UV.
