The story revolves around three roommates, journalist Tashi (Imran Khan), photographer Nitin Berry (Kunaal Roy Kapur) and cartoonist Arup (Vir Das), leading an unkempt and debt-ridden life in a shoddy apartment. Tashi's ditzy fiancée, Sonia (Shenaz Treasurywala), is an air hostess who agrees to deliver a package for Vladimir Dragunsky (Kim Bodnia) to Somayajulu (Vijay Raaz), without realizing its contents or that Somayajulu is a gangster. Sonia asks Tashi to deliver the package. Tashi, in turn, asks Nitin to do so. But Nitin is unable to do so as he is suffering 'Delhi Belly'. Nitin hands Sonia's package to Arup for delivery to Somayajulu, along with a package containing his stool sample for delivery to Nitin's doctor. Arup mixes up the two bags. Somayajulu, furious, tortures Vladimir to find his package.

The plot of the movie is based on the original script of Jayant Pawar, which highlights the consequences of the atrocities towards the mill workers, totally ignored by the Government. The film is a story of a family of mill workers in Mumbai. It traces the birth of the politics of greed in Mumbai and exposes the collusion between the triumvirate of big business, the political establishment and the trade union leaders who ostensibly were charged with protecting the rights of the mill workers. In the two decades that followed, the entire landscape of Central Bombay was changed forever. Land became the currency of growth, and this began the systematic extinction of mills in Bombay. In a matter of just a few years, hundreds of thousands of workers lost their means of livelihood. Having worked in these mills from generation to generation, this was the only vocation that they knew.

Salman Khan is a Bollywood actor, who is on a live tour nearby Delhi, where his private jet has a crash. Luckily, he survives and in a lounge, he meets Katrina Kaif. She tells him a story, about six friends and their boss who all work together at a Mumbai-based call centre. They are instructed never to reveal their location, and speak with an American accent by a Boston-based company. There is Shyam (Sam) (Sharman Joshi), Priyanka (Gul Panag), Varun (Sohail Khan), Esha (Isha Koppikar), Radhika (Amrita Arora) and Military uncle (Sharat Saxena). Their boss, Subhash Bakshi (Dalip Tahil), attempts to further his career by plagiarising software, agrees to lay off 40% of the Indian workforce, and re-locate to Boston. One night, at a party, they all get drunk and decide to head back home since it was late. The six are on their way home, until they approach an accident, and are stuck on top of a cliff in their car, and if anybody tries to move, there would be chances that the car will lose its balance and fall off the cliff. They all remain calm, and when one of the friends tries to call someone, he finds his phone battery has run out, therefore, in rage he breaks his phone and throws it on the floor of the car. Later on, still trapped in the car, the broken phone receives a phone call; six surprised friends answer it, only to find the caller is God. God tells the six, they should keep calm, and just believe in themselves and all will be well. They do as they are told, and find that the fire brigade are here to save them. They all get saved. After the story, Khan asks the woman who she is, and she replies "if you believe in yourself, you will know the answer". She walks out of the lounge, and as Khan follows her, he seems to witness she has suddenly disappeared. He believes in himself, his jet gets fixed, and he flies back home.

Small-time crook, Champak Chaudhary alias 'Champ' (Arshad Warsi) forges passports, photographs, and deeds in Thailand. He is arrested by the police, tried and sentenced to six months in jail. This is where he meets a supposedly mute and deaf inmate, Raghu Sharma (Raghuvir Yadav), serving a life sentence for stealing diamonds. Champ finds out that Raghu is not deaf nor mute and offers him a share if he springs him out of prison; Champ agrees. As soon as Champ's term is over, he gets discharged and prepares forged release documents for Raghu and gets him 'legally' out of jail.

The movie begins with Amar (John Abraham) narrating the story of his family and himself. Amar is the son of Vidyadhar (Amitabh Bachchan) and Sumitra Patwardhan (Sharmila Tagore), a middle class couple in India. Amar lives and works in London and he sends some of his salary to them. One day Amar returns home with Jenny Mayer (Anusha Dandekar), his girlfriend, and announces his intention to get married to her. Initially reluctant, his parents are pleased and happy for the two. Debu (Rahul Bose) is a 32-year-old advertising agent, but has a serious problem in that he is still a virgin. He meets Baba Hindustani (Vijay Raaz) in a hotel who promises to cure him of the teasing he gets from being a virgin. He later meets Nitin Kapoor, a film-maker (Saurabh Shukla) who later films him as he works out in the gym and other physical activities. Unaware of what is happening, Kapoor edits the film in such a way to give the impression of Debu as having sex. The film is released and becomes a box office hit and Debu quickly becomes a sex symbol. He later meets Sonali Verma (Perizaad Zorabian), a journalist, who helps him through his difficulties and they later fall in love.
