Dr. Andrew Manson (Robert Donat) is an idealistic newly qualified Scottish doctor dedicated to treating Welsh miners suffering from tuberculosis in the Welsh mining village of Blaenely and apprentice to Dr. Page (Basil Gill). Initially, he is full of lofty scientific goals, though his purpose erodes when he later moves to London to treat rich hypochondriacs, where a chance encounter with a medical school chum Dr. Frederick Lawford (Rex Harrison) leads to his quiet seduction by an ethically challenged medical establishment. Christine (Rosalind Russell), his wife tries to set him back on the original path. Dr. Philip Denny (Ralph Richardson), Manson's best friend, dies at the hands of an incompetent, social-climbing surgeon.

Englishman A. J. Fothergill (Robert Donat) is recruited by Colonel Forrester (Laurence Hanray) to spy on Russia for the British government because he can speak the language fluently. As "Peter Ouranoff", he infiltrates a revolutionary group led by Axelstein (Basil Gill). The radicals try to blow up General Gregor Vladinoff (Herbert Lomas), the father of Alexandra (Marlene Dietrich). When the attempt fails, the would-be assassin is shot, but manages to reach Peter's apartment, where he dies. For his inadvertent involvement, Peter is sent to Siberia.

Rembrandt, au moment où l'histoire commence, est un peintre adulé, sollicité par les plus riches, mais dont le plus grand plaisir consiste à réaliser des portraits de son épouse Saskia. Celle-ci, cependant, tombe brusquement malade et meurt, laissant le peintre désemparé. Plutôt que d'assister aux funérailles de sa femme, il tâche de peindre d'elle au moins un dernier portrait, tant que son image ne s'est pas encore tout à fait éteinte dans son souvenir ; à partir de cet événement tragique, la vie du peintre, progressivement, va se désagréger…
