Davis Okoye est un primatologue qui préfère se tenir à l'écart des hommes. Il partage un lien inébranlable avec George, un gorille albinos extraordinairement intelligent, qu'il côtoie depuis la naissance de l'animal. Leur relation va cependant être perturbée par une expérience génétique ratée (CRISPR) qui transforme George en un monstre furieux. Deux autres prédateurs (un loup et un crocodile) ont également subi les modifications génétiques. Ces créatures vont alors tout ravager sur leur passage dans la ville de Chicago. It has been four years since the events of the first movie. Seventeen-year-old Sean Anderson (Josh Hutcherson) is caught by the police after a brief chase on his dirtbike which ended with him driving into a swimming pool while trying to evade them. Minutes later, his stepfather Hank (Dwayne Johnson) arrives where a police officer (Stephen Caudill) who is friends with Hank tells him that Sean had broken into a remote satellite research center. The police officer also tells Hank that he has talked Mr. and Mrs. McGillicutty (the owners of the swimming pool) out of pressing charges. Hank takes Sean home where his mother Elizabeth (Kristin Davis) is not pleased with his actions or the fact that he and Hank don't get along well.

At a satellite base in Northern Germany, a worker named Freidrich, who is delivering secret codes finds a Cocker Spaniel puppy outside his office. He takes the puppy inside and shows it to his Bloodhound Rex. Rex senses that something is wrong with the dog and barks at it, making the worker take him outside. Both get locked out of the office. Rex looks through the window and sees the puppy taking pictures of the secret codes and revealing itself to be an evil female hairless Sphynx cat named Kitty Galore in disguise. Rex turns out to be a dog agent and reports to HQ.

Evelyn then tries to resurrect herself through suicide, after reading a book entitled How to Kill Yourself Back to Life. Her suicide attempts fail; part of the problem is that she wastes her bullets killing birds instead of herself, and she swings on her rope instead of hanging herself. Eventually, one boy (Close) is killed in a bus accident and becomes a living dead friend for her.
