To celebrate his upcoming marriage to Tracy Garner (Sasha Barrese), Doug Billings (Justin Bartha) travels to Las Vegas with his best friends Phil Wenneck (Bradley Cooper) and Stu Price (Ed Helms), as well as Tracy's brother and Doug's future brother-in-law Alan (Zach Galifianakis), in Doug's future father-in-law's vintage Mercedes-Benz. They stay at Caesars Palace, where they relax in the room before celebrating with a few drinks on the hotel rooftop. The next morning, Phil, Stu, and Alan awaken to find they have no memory of the previous night, and Doug is nowhere to be found. Stu is missing a tooth, their hotel suite is in disarray, a tiger is in their bathroom, a chicken in their living room, and a baby is in the closet, whom they name "Carlos". They find Doug's mattress impaled on a statue outside of their hotel and when they ask for their Mercedes, the valet delivers an LVPD police cruiser.

Edison (Carrot Top) is a poor, failed inventor and surf bum, but soon runs into and befriends wealthy business magnate Armand McMillan (Jack Warden). When Armand passes away shortly thereafter, he leaves Edison the majority of shares in his large invention corporation, and leaves his jealous nephew, his only living relative, Bradford (Larry Miller) only a surfboard. Bradford attempts to derail Edison's success by stealing his formula for glow in the dark, which had not previously existed in the reality of this movie, and sharing it with a consumer who covers himself in it and alleges that Edison's prize invention—a portable TV/TV dinner combo—leaks radiation. When Edison reveals to all that radiation causes sickness and death—but not a literal glowing—and that he had invented the formula for glow in the dark, the company is saved and Edison appoints beau Natalie Stockwell (Courtney Thorne-Smith) to run the company.

Some years after the previous film, Dennis Mitchell (Justin Cooper) is worse than ever. At the beginning of the movie, he goes over to Mr. Wilson's (Don Rickles) house to offer him one of several gifts for his birthday. These include lizards, snakes, bugs, and other creatures. This ordeal ends with Mr. Wilson unintentionally riding down a flight of stairs in Dennis' red wagon and accidentally getting his birthday cake thrown in his face by Martha (Betty White). Soon after this incident, Mr. Johnson, Dennis' grandfather and Alice's father (George Kennedy), shows up and announces that he is moving in with the Mitchells. Dennis starts spending more time with him than his annoyed neighbor. Mr. Wilson, upset that he's getting older, gets tricked by two con men (Brian Doyle-Murray and Carrot Top) who try to talk him into buying a "rare" root used to make tea to make people younger.
